摘  要:目的:通过改良一步酶消化法,快速便捷地培养人头皮毛乳头细胞。方法:采用一步酶消化法培养人头皮毛乳头细胞,并鉴定培养的人头皮毛乳头细胞。结果:通过镜下观察,发现一步酶消化法与改良二步酶消化法获得人头皮毛乳头细胞的有效率相当;相比传统方法,耗时更少,操作更便捷。培养的毛乳头细胞冻存复苏效果佳。结论:一步酶消化法可有效及高效地培养人头部毛乳头细胞。Objective: To improve the one-step enzyme digestion method, making the in vitro isolation and culture of human scalp dermal papilla cells more rapid and efficient. Methods: The improved one-step enzyme digestion method was employed to culture human dermal papilla cells which were then identified by alpha-SMA immunofluorescent staining. Results:Microscopic observation revealed that the efficiency of obtaining human dermal papilla cells by the improved one-stop enzyme digestion method was comparable to that of the improved two-step method. Compared with the conventional methods, human workload and operating time were reduced significantly. The cultured dermal papilla cells had a good recovery after cryopreservation. Conclusion: The improved one-step enzyme digestion method can culture human dermal papilla cells efficiently.
