摘  要:金属有机骨架(metal-organic framework,MOF)是中心金属离子或金属离子簇与有机配体通过配位作用自组装形成的结构规则的有机无机杂化晶体材料。MOF具有巨大的比表面积、极高的孔隙率以及尺寸形状可调节等特点,而且制备方法简单,引起医药领域的广泛关注,开始成为新一类具有应用前景的药物载体。然而,其组成成分的潜在毒性限制了MOF在生物医药领域的应用。近年来,有研究表明由γ-环糊精和钾离子构成的环糊精金属有机骨架(γ-cyclodextrin metal-organic framework, CD-MOF)组成成分安全无毒,具有良好的生物相容性,可解决MOF在药物递送时实际应用的不足,还可有效改善客体药物分子的稳定性,提高疗效,是极具潜力的药物载体。本文将从CD-MOF的合成与安全性、应用于药物载体的优势和局限进行综述。Metal-organic framework(MOF) is structurally regular organic-inorganic hybrid crystal material constructed by metal ions or metal ion clusters and organic ligands. MOF has attracted attention of biomedical field and would be a new class of drug carrier with promising applications due to its large specific surface area, extremely high porosity, and adjustable size and shape. However, the application of MOF was limited by its potential toxicity. Recent studies have shown that the biocompatible cyclodextrin metal-organic framework(CD-MOF) which is composed of γ-cyclodextrin and potassium ions has become a promising drug carrier because of its safety and non-toxicity. It can overcome the drawbacks in practical application of MOF in drug delivery, and can effectively improve the stability of drug molecules. This article discusses the synthesis, safety, advantages and limitations of CD-MOF as drug carrier.
