摘 要:为明确石河子市宿根福禄考(Phlox paniculata L.)斑枯病病原菌种类及其生物学特性。将病株叶片采用组织分离法进行分离、纯化获得单孢菌株。通过致病性测定、形态特征观察及分子生物学方法进行病原鉴定,室内测定了病原菌的生物学特性。结果表明,供试菌株对宿根福禄考幼苗均有致病性,菌株的分生孢子、分生孢子器、厚垣孢子的形态及不同培养基上的培养特性均与已报道的福禄考壳针孢(Septoria phlogis)相似。供试菌株rDNA-ITS区、β-微管蛋白及延长因子基因序列与S.phlogis同源性高达97.68%~100%;多基因联合构建系统发育树显示供试菌株与福禄考壳针孢(S.phlogis)均在同一分枝上。菌株最适生长温度和产孢温度分别为25℃和20℃;最适生长和产孢pH值为7.0。光暗交替有利于菌丝生长,连续黑暗有利于菌株产孢。将引起石河子市宿根福禄考斑枯病的病原鉴定为福禄考壳针孢(Septoria phlogis)。该病原菌生长和产孢适宜温度偏低,该病害为低温病害。In order to determine the pathogen and biological characteristics of leaf spot of Phlox paniculata L.in Shihezi,the diseased leaves were separated and purified by tissue separation to obtain single spore strain.After that pathogen identification was carried out by means of pathogenicity determination,morphological characteristics observation and molecular biological method,and the biological characteristics of pathogenic bacteria were determined indoors.The results showed that the tested strains were pathogenic to Phlox paniculata L.seedlings.The characteristics of conidia,conidiomata,chlamydospore and colonial morphology of the strains were consistent with the reported Septoria phlogis.The sequence of rDNA-ITS region,β-tubulin sequence and elongation factor sequence of the tested strain were 97.68%-100%homologous to S.phlogis;and the phylogenetic tree constructed by multiple genes showed that the tested strain and S.phlogis were all on the same branch.The biological characteristics showed that the optimal growth temperature and sporulation temperature of the pathogen were 25℃and 20℃,respectively,and the optimal growth and sporulation pH was 7.0.Alternating light and dark was conducive to the growth of mycelium,and continuous darkness was conducive to the spore production.Therefore,the pathogen causing leaf spot of Phlox paniculata L.was identified as S.phlogis in Shihezi.The suitable temperature for the growth and sporulation of the pathogen was low,so the disease belonged to a low temperature disease.
