摘 要:目的通过检测应激性高血压(stress-induced hypertention,SIH)大鼠肾组织中结缔组织生长因子(connective tissue growth factor,CTGF)、基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)、肿瘤坏死因子(tumor necrosis factor-α,TNF-α)的表达水平变化,初步探讨CTGF、MMP-9、TNF-α在应激性高血压大鼠肾脏损伤中的作用。方法应用复合应激建立SIH的动物模型,放射免疫法检测血浆中醛固酮(aldosterone,ALD)、血管紧张素II(Angiotensin II,Ang II)含量的变化;实时荧光定量PCR(RT-qPCR)及免疫组化检测大鼠肾组织中CTGF、MMP-9、TNF-α的表达变化。结果与正常对照组相比,SIH大鼠血浆中ALD、Ang II的含量明显升高(P<0.05),差异有统计学意义;SIH组大鼠肾脏CTGF、MMP-9、TNF-α的蛋白表达水平较正常对照组相比明显升高(P<0.05);CTGF的mRNA水平较正常对照组显著升高(P<0.05),但MMP-9无明显变化,而TNF-α较正常对照组相比是下调的。结论CTGF、MMP-9、TNF-α可能是SIH大鼠所至肾损伤的重要原因。Objective To detect the expression of CTGF,MMP-9 and TNF-αprotein in the renal tissue of stress-induced hypertention(SIH)rats and to explore the role of CTGF in renal injury of SIH rats.Methods The SIH rat model of was established by combined stress.The contents of aldosterone,angiotensin II in plasma were detected by radioimmunoassay,and the expressions of CTGF,MMP-9 and TNF-αin kidney were detected by RT-qPCR and immunohistochemistry.Results Compared with the normal group,the contents of aldosterone,angiotensin II in plasma of SIH rats were significantly higher than those in the normal group(P<0.05).The protein expression levels of renal CTGF,MMP-9 and TNF-αin the SIH group were significantly higher than those in the normal group(P<0.05);the mRNA level of CTGF of SIH rats was significantly higher than that of normal rats(P<0.05),but there was no significant change in MMP-9,while the TNF-αmRNA was down-regulated compared with the normal group.Conclusion CTGF,MMP-9 and TNF-αmay be an important cause of renal injury in SIH rats.
