摘  要:目的初步探讨应激性高血压(Stress-induced hypertension,SIH)大鼠血管组织内质网应激(Endoplasmic Reticulum Stress,ERS)与血管紧张素Ⅱ受体1型(AngiotensinⅡtype 1 receptor,AT1R)的关系。方法通过复合应激建立SIH模型。对大鼠胸主动脉石蜡包埋组织进行HE染色,观察大鼠胸主动脉壁中层平滑肌层的形态学变化;用实时荧光定量PCR(RT-qPCR)方法﹑免疫组织化学(IHC)方法及Western印迹(WB)对大鼠胸主动脉组织进行grp78和at1r基因mRNA表达水平以及GRP78和AT1R的蛋白表达水平的检测。结果与正常组相比,SIH模型组中膜肌层血管平滑肌细胞(vescular smooth muscle cell,VSMC)肥大、动脉血管壁增厚、细胞排列较紊乱、弹性膜弯曲及紊乱;SIH模型组大鼠胸主动脉血管组织中at1r基因mRNA表达水平较正常组显著升高(P<0.05);与正常组相比,GRP78和AT1R在SIH模型组大鼠胸主动脉组织中高表达(P<0.05)。结论SIH模型组大鼠血管组织中有ERS的发生,ERS下游信号分子通过激活转录因子诱导血管组织中ATIR的高表达,从而引起血压的升高。Objective To investigate the relationship between Endoplasmic Reticulum Stress(ERS)and AT1 receptor(AT1R)in rats with stress-induced hypertension(SIH).Methods The SIH model was established by combined stress.The morphological changes of the middle smooth muscle layer of the thoracic aortic wall were observed by HE staining.The expression levels of grp78,at1r gene and GRP78,AT1R protein of thoracic aortic tissues were detected by real-time fluorescence quantitative assay PCR(RT-qPCR),immunohistochemistry(IHC)and western blotting(WB).Results Compared with the normal group,VSMC hypertrophy,arterial wall thickening,cell arrangement disorder,elastic membrane bending and disorder appeared in the SIH model group;the mRNA expression level of at1r gene in the thoracic aortic vascular tissues of rats in the SIH model group was significantly higher than that in the normal group(P<0.05);Compared with the normal group,GRP78 and AT1R were highly expressed in the thoracic aorta in the model group(P<0.05).Conclusion ERS may occur in the vascular tissues of rats in the SIH model group.The downstream signal molecules of ERS induce the high expression of ATIR in the vascular tissues by activating transcription factors,so as to cause the increase of blood pressure.
