摘 要:目的观察芦荟苷(Barbaloin,BAR)对大鼠心肌缺血/再灌注性损伤(MIRI)的改善作用,并探索其保护机制。方法通过结扎冠状动脉左前降支30 min再灌注60 min的方法建立大鼠心肌缺血再灌注损伤(MI/R)模型,72只SD大鼠随机分为对照组,模型组,阳性对照组(地尔硫卓0.01 mg/kg预处理),BAR低浓度预处理组(10 mg/kg),BAR中浓度预处理组(50 mg/kg)和BAR高浓度预处理组(100 mg/kg),每组12只。比色法检测血清及心肌组织一氧化氮(NO)及总抗氧化能力(TAOC);Elisa法检测心肌组织丙二醛(MDA),超氧化物歧化酶(SOD),过氧化氢酶(CAT),谷胱甘肽过氧化物酶(GSH-px)含量;流式细胞仪检测心肌细胞凋亡率;western blot检测内皮型一氧化氮合酶(eNOS)磷酸化水平、蛋白激酶B(Akt)蛋白磷酸化水平以及凋亡相关蛋白caspase-3,Bax/Bcl-2表达水平。结果模型组大鼠血清及心肌组织NO、TAOC显著低于对照组(P<0.01),芦荟苷组在血清及心肌组织NO、TAOC表达水平显著高于模型组(P<0.05)。结果显示,模型组大鼠血清及心肌组织MDA含量均显著高于对照组(P<0.01),芦荟苷组中血清及心肌组织MDA含量显著下降(P<0.05);模型组大鼠血清及心肌组织SOD、CAT、GSH-px含量均显著低于对照组(P<0.01),芦荟苷组中血清及心肌组织SOD、CAT、GSH-px含量均显著提高(P<0.05)。模型组大鼠心肌细胞凋亡率明显高于对照组(P<0.01),芦荟苷低、中、高预处理组显著低于模型组,并呈剂量依赖趋势(P<0.05)。与对照组比较,模型组大鼠心肌组织Akt、eNOS蛋白磷酸化水平明显降低(P<0.01),与模型组比较,芦荟苷组心肌组织Akt、eNOS蛋白磷酸化水平显著升高(P<0.05)。与对照组比较,模型组大鼠心肌组织caspase-3、Bax蛋白表达显著提高(P<0.01),与模型组比较,芦荟苷组大鼠心肌组织caspase-3、Bax蛋白表达显著降低,并呈剂量依赖趋势(P<0.05);而模型组大鼠心肌组织Bcl-2蛋白表达显著低于对照组(P<0.01),芦荟苷�Objective To investigate the effects of barbaloin pretreatment on myocardial ischemia/reperfusion injury(MI/R)in rats,and to explore its action mechanism.Methods The rat models with MI/R were established by coronary artery ligation of the left anterior descending branch(30min/60min),and the 72 rats were divided into 6 groups:control group,model group,positive control group(diltiazem pretreatment 0.01mg/kg),low concentration pretreatment group(10mg/kg),medium concentration pretreatment group(50mg/kg)and high concentration pretreatment group(100mg/kg),with 12 rats in each group.After that th levels of nitric oxide(NO)and total antioxidant capacity(TAOC)in serums and myocardial tissue were detected by colorimetry method.The levels of malondialdehyde(MDA),superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-px)were detected by ELISA.Moreover the myocardial apoptosis rate was detected by flow cytometry.In addition thelevels of endothelial nitric oxide synthase(eNOS)phosphorylation level,protein kinase B(Akt),caspase-3 and Bax/Bcl-2 were detected by Western Blot.Results The levels of NO and TAOC in serums and myocardial tissue in model groups were significantly lower than those in control group(P<0.01),which in barbaloin groups were significantly higher than those in model group(P<0.05).The levels of MDA in serums and myocardial tissue in model groups were significantly higher than those in control group(P<0.01),which in barbaloin groups were significantly decreased(P<0.05).Moreover the levels of SOD,CAT and GSH-px in serums and myocardial tissue in model group were significantly lower than those in control group(P<0.01),which in barbaloin groups were significantly increased.In addition myocardial apoptosis rate in model group was significantly lower than that in control group(P<0.01),which in barbaloin low concentration pretreatment group,medium concentration pretreatment group and high concentration pretreatment group was significantly lower than that in model group,in a doae-dependent manner(P<0.05).Be
