摘 要:目的探讨盐酸羟考酮对异丙肾上腺素致心肌损伤的保护作用及机制。方法用皮下注射异丙肾上腺素的方法建立小鼠心肌损伤模型。按照体重将雄性昆明种小鼠随机分为4组:正常组、模型组、普萘洛尔组和盐酸羟考酮组,每组10只。普萘洛尔组予以普萘洛尔30 mg·kg^-1灌胃,盐酸羟考酮组予以盐酸羟考酮30μg·kg^-1灌胃,正常组和模型组小鼠给予等剂量0.9%Na Cl灌胃,均连续灌胃3 d。用分光光度法测定各组小鼠血清中丙二醛(MDA)和谷甘肽过氧化物酶(GSHpx)水平;用蛋白质印迹法测定烟酰胺腺嘌呤二核苷酸磷酸(NADPH)氧化酶亚基的相对表达量。结果正常组、模型组、普萘洛尔组和盐酸羟考酮组的MDA分别为(4.56±0.61),(7.45±1.33),(6.09±1.12)和(6.45±0.98)nmol·mL^-1;这4组GSHpx水平分别为(654.6±109.4),(342.6±78.7),(466.5±91.1)和(487.8±68.7)U·mL^-1;这4组的P22subunit/β-actin相对表达量分别为0.30±0.06,0.51±0.12,0.45±0.08和0.49±0.11;这4组的P47subunit/β-actin相对表达量分别为0.26±0.05,0.42±0.07,0.34±0.06和0.38±0.08;这4组的P67subunit/β-actin相对表达量分别为0.28±0.06,0.50±0.12,0.43±0.08和0.37±0.09。上述指标:模型组与正常组比较,差异均有统计学意义(均P<0.01);普萘洛尔组和盐酸羟考酮组与模型组比较,差异均有统计学意义(均P<0.05);普萘洛尔组与盐酸羟考酮组比较,差异均有统计学意义(均P<0.05)。结论盐酸羟考酮能通过抑制氧化应激对异丙肾上腺素致心肌损伤发挥保护作用。Objective To explore the protective effect and mechanism of oxycodone hydrochloride on myocardial injury mice induced by isoproterenol.Methods The mice model of myocardial injury was established by subcutaneous injection of isoproterenol.Male Kunming mice were randomly divided into four groups:normal group,model group,propranolol group(propranolol 30 mg·kg^-1)and oxycodone hydrochloride group(oxycodone 30μg·kg^-1),normal group and model group were given equal doses of normal saline,for 3 d.The levels of malondialdehyde(MDA),glutathione peroxidase(GSHpx)in plasma of mice were measured by spectrophotometry.And relative expression of the myocardial reduced coenzyme II(NADPH)oxidase were measured by Western blot.Results The MDA in the serum of normal group,model group,propranolol group and oxycodone hydrochloride group were(4.56±0.61),(7.45±1.33),(6.09±1.12)and(6.45±0.98)nmol·mL-1 respectively;the GSHpx levels of the four groups were(654.6±109.4),(342.6±78.7),(466.5±91.1)and(487.8±68.7)U·mL-1;the relative expression of the P22 subunit/β-actin levels of the four groups were 0.30±0.06,0.51±0.12,0.45±0.08 and 0.49±0.11,respectively;the relative expression of the P47 subunit/β-actin levels of the four groups were 0.26±0.05,0.42±0.07,0.34±0.06 and0.38±0.08,respectively;the relative expression of the P67 subunit/β-actin levels of the four groups were0.28±0.06,0.50±0.12,0.43±0.08 and 0.37±0.09,respectively.There were significant differences of the factors between model group and normal group;there were significant differences of the factors between propranolol group,oxycodone hydrochloride group and model group(all P<0.05);there were significant differences of the factors between propranolol group and oxycodone hydrochloride group(all P<0.05).Conclusion Oxycodone hydrochloride can protect myocardial injury induced by isoproterenol by inhibiting oxidative stress.
