摘 要:目的研究三七总皂苷对局部脑缺血大鼠的神经保护作用。方法按照随机数字法将雄性SD大鼠分为4组:正常组、模型组,阿替普酶组和联合组,各20只。模型组、阿替普酶组和联合组的大鼠制作局部脑缺血模型。正常组、模型组的大鼠均不给予任何治疗,阿替普酶组大鼠在造模后3 h给予静脉注射阿替普酶(2 mL·kg^-1),联合组大鼠在阿替普酶组的基础上给予三七总皂苷(20mg·kg^-1)。用Garcia评分法评价大鼠术后24 h的神经功能评分,用邻苯三酚自氧化测定丙二醛含量,用硫代巴比妥酸法测定超氧化物歧化酶(SOD)含量。结果正常组、模型组、阿替普酶组和联合组的神经功能评分分别为(17.12±0.63),(10.01±1.78),(12.41±1.07)和(13.52±1.25)分;这4组的MDA分别为(301.5±23.5),(513.2±37.8),(470.8±39.2)和(410.3±30.7)μmo L·mg^-1;这4组SOD分别为(0.50±0.04),(0.36±0.04),(0.38±0.04)和(0.42±0.03)U·mg^-1。上述指标:正常组与模型组相比、或者阿替普酶组和联合组与模型组相比,差异均有统计学意义(均P<0.05)。结论用三七总皂苷干预局部脑缺血的大鼠模型,可以提升阿替普酶的疗效,保护神经功能,抑制炎性反应和氧化应激反应。Objective to investigate the neuroprotective effect of total saponins from Panax notoginseng(panax notoginseng)in cerebral ischemia rats.Methods Male SD rats were selected and randomly divided into 4 groups:normal group,model group,alteplase group and joint group.Rats in model group,alteplase group and joint group were used to make models of cerebral ischemia.Rats in the normal and model groups were not given any treatment.Rats in the alteplase group were given intravenous alteplase injection(2 mL·kg^-1)3 h after the model preparation.Rats in joint group were given total saponins of Panax notoginseng(20 mg·kg^-1)basis on the alteplase group.Garcia scoring method was used to evaluate the neurological function score of rats 24 h after the operation.The content of malondialdehyde(MDA)was determined by catechol self-oxidation and superoxide dismutase(SOD)was determined by thiobarbituric acid.Results The neurological function scores in normal group,model group,alteplase group and joint group were(17.12±0.63),(10.01±1.78),(12.41±1.07)and(13.52±1.25)point,respectively;MDA in the four groups were(301.5±23.5),(513.2±37.8),(470.8±39.2)and(410.3±30.7)μmoL·mg^-1,respectively;SOD levels in the four groups were(0.50±0.04),(0.36±0.04),(0.38±0.04)and(0.42±0.03)U·mg^-1,respectively.There were statistically significant differences of the factors between normal group and model group,or between alteplase group and joint group with model group(all P<0.05).Conclusion The intervention of total saponins of Panax notoginseng in rats with cerebral ischemia can improve the therapeutic effect of alteplase,protect nerve function,and inhibit inflammatory response and oxidative stress response.
