摘  要:目的探讨端粒酶抑制因子X1(PinX1)对皮肤鳞状细胞癌细胞生长与凋亡的影响。方法将SCL-1细胞分成3组:空白对照组(空白对照细胞)、阴性对照组(转染阴性对照载体)和过表达组(转染PinX1过表达载体)。以噻唑蓝法检测细胞增殖能力(OD值),以流式细胞术检测细胞凋亡,以反转录-聚合酶链反应测定SCL-1细胞中PinX1基因表达水平,以蛋白质印迹检测Notch受体胞内结构域1(NICD1)、Notch受体胞内结构域2(NICD2)、发状分裂相关增强子1(HES1)和端粒酶逆转录酶(hTERT)和PinX1蛋白表达。结果空白对照组、阴性对照组和过表达组细胞中的PinX1基因表达水平分别为1.00±0.12,1.04±0.07和3.25±0.21;这3组细胞的增殖能力分别为1.26±0.10,1.24±0.12和0.85±0.09;这3组细胞的凋亡率分别为(9.39±0.93)%,(9.59±1.21)%和(27.88±2.48)%;这3组细胞的NICD1蛋白水平分别为0.62±0.05,0.60±0.07和1.15±0.14;这3组细胞中的NICD2蛋白水平分别为0.55±0.04,0.57±0.06和1.24±0.12;这3组细胞中的HES1蛋白水平分别为0.53±0.07,0.54±0.06和0.91±0.09;这3组细胞中的hTERT蛋白水平分别为0.51±0.04,0.52±0.05和0.17±0.03。上述指标:过表达组与阴性对照组比较,差异均有统计学意义(均P<0.05)。结论PinX1可能通过激活Notch信号通路,抑制皮肤鳞状细胞癌细胞生长和诱导细胞凋亡。Objective To investigate the effect of the Pin2/TRF1-interacting protein X1(PinX1)on the growth and apoptosis of Squamous cell carcinoma of skin.Methods SCL-1 cells were divided into three groups:blank control group(blank control),negative control group(transfection control empty vector),overexpression group(transfection PinX1 overexpression vector).The expression levels of PinX1 mRNA and protein in cells were detected by reverse transcription-PCR and Western blot,respectively.Cell proliferation was detected by methylthiazolyldiphenyl-tetrazolium bromide.Apoptosis rate was detected by flow cytometry.The expression of Notch signaling pathway connected protein Notch intra-cellular domain1(NICD1),Notch intra-cellular domain2(NICD2),hairy and enhancer of split 1(HES1),human telomerase reverse transcriptase(hTERT)were detected by Western blot.Results The expression levels of PinX1 mRNA in blank control group negative control group,overexpression group were 1.00±0.12,1.04±0.07,3.25±0.21;the ability of cell proliferation in the three groups were 1.26±0.10,1.24±0.12,0.85±0.09;the apoptotic rate in the three groups were(9.39±0.93)%,(9.59±1.21)%,(27.88±2.48)%;the levels of NICD1 protein in the three groups were0.62±0.05,0.60±0.07 and 1.15±0.14;the levels of NICD2 protein in the three groups were 0.55±0.04,0.57±0.06 and 1.24±0.12;the levels of HES1 protein in the three groups were 0.53±0.07,0.54±0.06 and0.91±0.09;the levels of hTERT protein in the three groups were 0.51±0.04,0.52±0.05,0.17±0.03.Comparison between overexpression group and negative control group,the difference of the factors were significantly(all P<0.05).Conclusion PinX1 inhibit the growth of Squamous cell carcinoma cells and induce apoptosis by activating the Notch signaling pathway.
