摘  要:目的研究甘草酸对特应性皮炎(AD)小鼠皮肤屏障功能及皮损组织cathelin相关抗菌肽(CRAMP)、胸腺基质淋巴生成素(TSLP)表达的影响。方法将C57BL/6小鼠80只,随机分为空白组(n=20)、模型组(n=20)、对照组(n=20)及实验组(n=20)。除空白组外,其余3组小鼠均用背部涂抹0.5%对二硝基氟苯(DNFB)混合液的方法建立特应性皮炎小鼠模型。造模成功后,实验组小鼠灌胃甘草酸50 mg·kg^-1·d^-1,对照组小鼠灌胃波尼松龙10 mg·kg^-1·d^-1,空白组与模型组灌胃等量生理盐水,连续干预21 d。用皮肤测试仪检测经皮水分丢失量(TEWL)及皮肤厚度;用酶联免疫吸附(ELISA)法检测血清免疫球蛋白E(IgE)、干扰素-γ(IFN-γ)和白细胞介素-4(IL-4)水平;用苏木精-伊红(HE)染色观察小鼠皮损组织病理变化;用免疫组化法检测皮损组织CRAMP、TSLP蛋白表达。结果空白组、模型组、实验组及对照组小鼠TEWL分别为(4.83±0.46),(32.64±4.37),(16.63±0.86),(11.34±0.59)g·m^-2·h^-1;皮肤厚度分别为(0.31±0.04),(1.22±0.23),(0.93±0.11),(0.49±0.07)mm;各组小鼠皮损组织CRAMP蛋白阳性细胞表达百分比分别为(94.32±2.73)%,(21.37±1.68)%,(56.64±4.22)%,(82.51±3.96)%;TSLP蛋白阳性细胞表达百分比分别为(18.62±3.34)%,(87.46±2.87)%,(69.23±3.48)%,(46.96±4.11)%。与空白组相比,模型组小鼠TEWL、皮肤厚度、血清IgE、IFN-γ、IL-4水平及皮损组织TSLP蛋白阳性细胞表达百分比显著升高,且实验组低于模型组,对照组低于实验组;皮损组织CRAMP蛋白阳性细胞表达百分比显著降低,且实验组高于模型组,对照组高于实验组(均P<0.05)。结论甘草酸可调节特应性皮炎小鼠免疫功能,抑制炎性反应,同时上调皮损组织CRAMP蛋白表达,下调TSLP蛋白表达,进而保护皮肤屏障功能。Objective To investigate the effects of glycyrrhizin on the skin barrier function and the expression of cathelin-associated antimicrobial peptide(CRAMP)and thymic stromal lymphopoietin(TSLP)in skin lesions tissues in atopic dermatitis(AD)mice.Methods Eighty C57 BL/6 mice were randomly divided into blank group(n=20),model group(n=20),control group(n=20)and test group(n=20).Except blank group,other three groups of mice were implanted with 0.5%solution of dinitrofluorobenzene(DNFB)on the back to establish an AD mouse model.After successful modeling,the mice in test group were gavage with glycyrrhizic acid 50 mg·kg^-1·d^-1,the rats in control group were gavage with prednisolone 10 mg·kg^-1·d^-1,blank group and model group were gavage with the same amount of normal saline for21 d.Transdermal water loss(TEWL)and skin thickness were measured by skin tester;serum immunoglobulin E(IgE),interferon-γ(IFN-γ)and interleukin-4(IL-4)levels were measured by enzyme-linked immunosorbent assay(ELISA);hematoxylin-eosin(HE)staining was used to observe the pathological changes of mouse skin lesions tissues;the expression of CRAMP and TSLP protein in skin lesions tissues were detected by immunohistochemistry method.Results The TEWL of blank group,model group,test group and control group were(4.83±0.46),(32.64±4.37),(16.63±0.86)and(11.34±0.59)g·m^-2·h^-1;the skin thicknesses were(0.31±0.04),(1.22±0.23),(0.93±0.11)and(0.49±0.07)mm;the percentages of CRAMP protein positive cells in skin lesions tissues were(94.32±2.73)%,(21.37±1.68)%,(56.64±4.22)%and(82.51±3.96)%;the percentages of TSLP protein positive cells were(18.62±3.34)%,(87.46±2.87)%,(69.23±3.48)%and(46.96±4.11)%.Compared with blank group,the TEWL,skin thickness,serum IgE,IFN-γ,IL-4 levels and the percentage of TSLP protein positive cells in skin lesions tissues significantly increased in model group,and test group were lower than model group,and control group were lower than test group;the percentage of expression of CRAMP protein positive cells in skin le
