摘 要:目的建立高效液相色谱(HPLC)法测定不同产地紫锥菊提取物中7种酚酸类成分的含量;层次聚类分析法(HCA)对其进行统计归类,为质量的一致性和合理应用提供依据。方法色谱柱为Inertsil C18色谱柱(250 mm×4.6 mm,5μm);流动相:0.1%磷酸水(A)-甲醇(B),梯度洗脱;流速0.8 mL·min-1;柱温为30℃;检测波长280 nm;进样量为10μL。对不同产地紫锥菊中酚酸含量进行聚类分析。结果 HPLC法可用于准确测定紫锥菊中酚酸类成分的含量,7种酚酸类成分在该色谱条件下有良好的分离度,在相应的线性范围内呈现良好的线性关系,平均回收率均在98.41%~101.9%,RSD<3.0%。结论建立的测定方法简便、准确,结合层次聚类分析可用于紫锥菊及其提取物的质量控制。Objective To establish a method for the determination of seven phenolic acids in Echinacea purpurea extracts from different producing areas by high performance liquid chromatography(HPLC),and to classify them by hierarchical clustering analysis(HCA),for the aim of providing basis for quality consistency and rational application.Methods The analysis was performed on an Inertsil C 18 column(250 mm×4.6 mm,5μm),mobile phase of 0.1%phosphoric acid water(A)-methanol(B)in a linear gradient mode.The flow rate was 0.8 mL·min-1.The detector wavelength was 280 nm and the column temperature was 30℃.The injection volume was 10μL.The content of phenolic acids in Echinacea purpurea from different areas was analyzed by cluster analysis.Results The content of phenolic acids in Echinacea purpurea could be accurately determined by HPLC.Seven phenolic acids had good separation resolution under this chromatographic condition,and showed good linear relationship in the corresponding linear range.The average recovery was in the range of from 98.41%to 101.9%,RSD<3.0%.Conclusion The established method is simple and accurate,which can be used for the quality control of Echinacea purpurea and its extracts by combining with hierarchical cluster analysis.
