摘  要:目的观察电针干预对放射线照射小鼠海马区突触结构及突触蛋白-1的影响。方法 40只C57BL/6J小鼠(30日龄)随机分成对照组、放射线照射组(8Gy)、电针穴位组、电针非穴位组,每组10只。水迷宫方法检测各组小鼠空间学习记忆功能,免疫组化方法检测各组小鼠海马区突触超微结构,分子生物学方法检测各组小鼠海马区突触蛋白-1基因及蛋白表达变化。结果与对照组比较,放射线照射组小鼠寻找平台潜伏期呈现增加趋势,其中在第2天、第3天、第4天与第5天差异具有统计学意义(P<0.01,P<0.05,P<0.01,P<0.001),穿越目的象限次数和在目的象限停留时间均显著减少(P<0.01,P<0.01)。与放射线照射组比较,电针穴位组小鼠潜伏期显著缩短(P<0.01,P<0.01,P<0.001),穿越目的象限次数及停留时间显著增加(P<0.05)。与对照组比较,放射线照射组小鼠海马区突触间隙及突触后致密区厚度均显著减小(P<0.01,P<0.05),而电针穴位干预后则显著增加(P<0.05,P<0.05)。与对照组比较,放射线照射组小鼠海马区突触蛋白-1基因及蛋白表达均显著下降(P<0.01,P<0.05)。与放射线照射组比较,电针穴位组突触蛋白-1基因及蛋白表达均显著上调(P<0.05,P<0.01)。电针非穴位组无上述影响。结论电针穴位干预可一定程度改善放射线照射所致的突触结构及功能的损伤,从整体促进学习记忆行为的改变。Objective To investigate the effect of electroacupuncture intervention on hippocampal synaptic structures and synapsin-1 in irradiated mice.Method Forty C57BL/6J mice(30 days old)were randomized to control,irradiation,point elecroacupuncture and non-point elecroacupuncture groups,10 rats each.Spatial learning and memory function was assessed using the Morris water maze in every group of mice.Hippocampal synaptic structures were observed by immunohistochemical method in every group of mice.The expressions of synapsin-1 and protein in the hippocampus were determined by molecular biology methods.Result Platform-finding latency tended to increase and had a statistically significant difference at day two,three,four and five(P<0.01,P<0.05,P<0.01,P<0.001)and the number of crossing and time spent in the target quadrant decreased significantly(P<0.01,P<0.01)in the irradiation group of mice compared with the control group.The latency shortened significantly(P<0.01,P<0.01,P<0.001)and the number of crossing and time spent in the target quadrant increased significantly(P<0.05)in the point elecroacupuncture group of mice compared with the irradiation group.Compared with the control group,hippocampal synaptic cleft and postsynaptic dense area thickness decreased significantly in the irradiation group of mice(P<0.01,P<0.05)and increased significantly after point electroacupuncture intervention(P<0.05,P<0.05).The expressions of synapsin-1 and protein in the hippocampus decreased significantly in the irradiation group of mice compared with the control group(P<0.01,P<0.05)and were up-regulated significantly in the point elecroacupuncture group compared with the irradiation group(P<0.05,P<0.01).There were no the above effects in the non-point elecroacupuncture group.Conclusion Point electroacupuncture intervention can reduce irradiation-caused injuries in synaptic structures and function to a certain extent and promote changes in learning and memory behaviors in a holistic manner.
