摘  要:为了分析鲜人参膏(fresh ginseng paste,FGP)对环磷酰胺(cyclophosphamide,CTX)诱导的免疫抑制小鼠免疫功能的影响,本研究选取BALB/c小鼠脾淋巴细胞,采用5种浓度(12.5、25.0、50.0、100.0和200.0 g/mL)的FGP刺激淋巴细胞的体外转化。另取ICR小鼠50只,随机分为5组:正常对照组(N)和免疫抑制模型组(CTX)给予生理盐水,免疫抑制组给予FGP,即FGP-L、FGP-M和FGPH 3个给药组分别给予150、300和600 mg/kg的FGP。模型组和给药组在第1、2、3、9、16、23和30天按80 mg/kg给小鼠腹腔注射环磷酰胺,测定FGP对BALB/c小鼠淋巴细胞体外转化的影响。ICR小鼠给药30 d后处死,测定胸腺指数和脾脏指数,评价FGP对脾淋巴细胞转化率的影响及血清中白细胞介素-2(IL-2)及干扰素-γ(IFN-γ)的含量。结果表明,FGP均能显著提高免疫抑制小鼠的胸腺指数和脾脏指数(P<0.05),可以明显促进体外脾淋巴细胞的转化。同时,FGP均能显著促进脾细胞的自然转化。其次,FGP能够显著提高小鼠血清中IL-2和IFN-的含量(P<0.01,P<0.05)。研究结果提示,FGP具有很强的免疫增强活性,并能拮抗环磷酰胺的免疫抑制作用。This work aims to study the effect of fresh ginseng paste(FGP) on the immune function of immunosuppressed mice induced by cyclophosphamide(CTX). Spleen lymphocytes of BALB/c mice were taken and stimulated to transform in vitro with 5 concentrations of FGP(12.5, 25.0, 50.0, 100.0, 200.0 g/mL). Another 50 ICR mice were randomly divided into 5 groups. Normal control group(N) and immunosuppressive model group(CTX) were given saline, and the administration groups were treated with different concentrations of FGP,which named FGP-L, FGP-M and FGP-H. The model group and the administration group were intraperitoneally injected with cyclophosphamide at 80 mg/kg on the 1st, 2nd, 3th, 9th, 16th, 23th, and 30th days. The effects of FGP on the transformation in vitro of lymphocytes in mice was measured. All ICR mice were sacrificed 30 days after administration, and thymus and spleen indices were measured. The effects of FGP on the transformation rate of spleen lymphocytes and the levels of interleukin-2(IL-2) and interferon-γ(IFN-γ) in serum were evaluated. FGP can significantly increase the thymus and spleen indices of immunosuppressed mice(P < 0.05), and it can significantly promote spleen lymphocyte transformation in vitro. In addition, FGP can significantly promote the natural transformation of spleen cells. Finally,FGP can significantly increase the serum levels of IL-2 and IFN-(P < 0.01, P < 0.05). FGP has strong immune-enhancing activity and it can antagonize the immunosuppressive effect of cyclophosphamide.
