摘 要:为探究VcPCNA基因在蓝莓果实发育过程中可能的调控机制,以南高丛蓝莓中果品种‘奥尼尔’与小果品种‘蓝雨’为实验材料,分离蓝莓VcPCNA基因全长cDNA,应用实时荧光定量PCR(qPCR)技术检测VcPCNA基因在花芽膨大与果实发育进程中的表达规律.结果表明:VcPCNA基因开放阅读框全长795 bp,包含保守的PIN00057功能域,以及可与其他蛋白互作的结合基序PIP-box,共编码264个氨基酸残基,相对分子质量为29.3 kDa;VcPCNA基因在‘蓝雨’花芽膨大期间中表达持续下降,而在‘奥尼尔’花芽中呈先上升后下降的趋势,且在开花前期VcPCNA表达量均处于较低水平;在果实发育进程中,‘奥尼尔’与‘蓝雨’VcPCNA基因表达模式相似,在开花期维持较低水平,在细胞快速分裂的S1期迅速升高至峰值后降低.结果表明,VcPCNA基因是动植物中保守的细胞增殖相关基因,其表达与蓝莓花芽、果实的发育进程密切相关,但其调控机制仍需深入研究.Proliferating cell nuclear antigen(PCNA)was a highly conserved DNA polymerase auxilins in eukaryotes.Medium-size fruit blueberry cultivar′O′Neal′and small-size fruits cultivar′Bluerain′were used to isolate the coding sequences of VcPCNA,and their relative expression patterns were detected during the flower bud and fruit development by quantitive real-time PCR(qPCR).The results showed that the open reading frame of VcPCNA gene was 795 bp,deduced VcPCNA contained a conserved PLN00057 domain,and a binding motif(PIP cassette)interacted with other proteins sequence.It encoded 264 amino acid residues,and its predicted molecular mass was 29.3 kDa.qPCR showed that the expression levels of VcPCNA gene was decreased continually during the′Bluerain′flower bud enlargement,while exhibited an"up and fall"pattern in the′O′Neal′flower buds.During the fruit development,the expression patterns of′O′Neal′and′Bluerain′VcPCNA were similar,which rose to the peak rapidly from stage S1,and decreased at stage S3 or S4.As to the second rapid fruit growth stage,VcPCNA mRNAs also exhibited an"up and fall"tendency.The results indicated that the VcPCNA gene was a conserved gene for cell proliferation and tissue development,however,its molecular regulation mechanism for blueberry fruit size remained unclear and needed to be further studied.
