复方中药提取液对家兔血液动力学效应影响的研究

摘　　要：研究旨在观察以丹参、三七和黄芪为方剂的中药提取液对家兔血液动力学效应的影响。25只家兔随机分为5组,Ⅰ组对照组、Ⅱ组模型组、Ⅲ组药物低剂量组(0.2 g/ml)、Ⅳ组药物中剂量组(0.4 g/ml)、V组药物高剂量组(0.8 g/ml)。除对照组外,其他家兔通过耳缘静脉注射20%甘露醇溶液建立模型。第4 d开始给药,7 d后进行抗凝血酶Ⅲ(AT-Ⅲ)、抗凝蛋白C(PC)、内皮素-1(ET-1)、血栓烷素(TXB2)、血小板因子4(PF4)和组织型纤溶酶原激活物(t-PA)指标检测。结果表明:造模后,与Ⅰ组相比,受试组AT-Ⅲ活性显著降低及TXB2活性显著升高(P<0.05);家兔耳缘静脉血管肉眼可明显观察到血管扩张,质地坚硬,呈暗红色,显示造模成功。给药7 d后,与Ⅱ组相比,Ⅳ、Ⅴ组的ET-1、TXB2和PF4的活性显著降低(P<0.05),AT-Ⅲ、PC、t-PA活性显著升高(P<0.05)。但在Ⅴ组给药过程中,家兔出现了不同程度的少食、精神萎靡及死亡等现象。综上所述,以丹参、三七和黄芪为方剂的中药提取液对家兔血液动力学效应有明显的改善效果,且药物浓度在0.4 g/ml时为最佳治疗剂量。The aim of the study was to explore that the effects of traditional Chinese medicine extracts of salvia miltiorrhiza combined with panaxnotoginseng and astragalus propinquus schischkin on haemodynamic effect of rabbits.25 rabbits were randomly divided into 5 groups.Ⅰ was control group;Ⅱ was model group;Ⅲ was extraction solution low dose group(0.2 g/ml);Ⅳ was extraction solution medium dose group(0.4 g/ml);Ⅴwas extraction solution high dose group(0.8 g/ml).In addition to the control group,other rabbits were injected with 20%mannitol solution to establish the model.The administration began on the 4th day,antithrombin Ⅲ(AT-Ⅲ),coagulant C(PC),endothelin-1(ET-1),thromboxane 2(TXB2),platelet factor 4(PF4)and tissue plasminogen activator(t-PA)were measured after administration for 7 days.The results showed that compared with group Ⅰ,AT-Ⅲ activity decreased and TXB2 activity increased significantly(P<0.05)in test group;the blood vessels of the ear vein of the rabbits were obviously observed to be dilatedwith a hard texture and dark red color,which indicated that the model was successful.Compared with group Ⅱ,the activity of ET-1,TXB2 and PF4 were down-regulation(P<0.05),and the activity of AT-Ⅲ,PC,t-PA were up-regulation significantly(P<0.05)in group Ⅳ and Ⅴ after administration for 7 days.However,during the administration of group Ⅴ,some phenomena such as low food intake,mental depression and death were observed in rabbits.In summary,salvia miltiorrhiza,panaxnotoginseng and astragalus propinquus schischkin had obvious therapeutic effects on rabbits haemodynamic effect,and the optimal therapeutic dose was 0.4 g/ml.
