摘　　要：目的地龙是巨蚓科动物参环毛蚓Pheretima aspergillum(E.Perrier)、通俗环毛蚓Pheretima vulgaris Chen、威廉环毛蚓Pheretima guillelmi(Michaelsen)或栉盲环毛蚓Pheretima pectinifera Michaelsen的干燥体.具有清热定惊,通络,平喘,利尿的功效.近年来,由于需求量大,出现地龙掺假的现象,于是采用分子生物学方法来鉴定地龙的真伪,同时建立一种鉴定地龙的方法.方法提取地龙粉末、地龙水提物、地龙醇提物、风湿祛痛胶囊的DNA,采用PCR方法得到PCR产物,并电泳鉴定其产物大小,鉴定地龙真伪,通过实验验证方法的稳定性.结果研究发现含有地龙的制剂的PCR产物均在260bp之间有单一条带,并摸索得出鉴定的条件为:95℃预变性5 min,35个循环(95℃变性30 s,58℃30 s,72℃30 s),72℃延伸5 min,经过验证具有稳定性.Earthworm is the dry body of the giant earthworm(e.Perrier),the common earthworm(Pheretima vulgaris Chen),the William earthworm(Pheretima guillelmi)(michaelsen)or the ctenoptera spectinifera michaelsen.It has the effect of clearing away heat,calming convulsion,dredging collaterals,relieving asthma and diuresis.In recent years,due to the large demand,the phenomenon of adulteration of earthworm appears,so the molecular biology method is used to identify the authenticity of earthworm,and a method of identification of earthworm is established.Methods the DNA of earthworm powder,earthworm water extract,earthworm alcohol extract and Fengshi Qutong capsule were extracted.The PCR products were obtained by PCR,and the size of the products was identified by electrophoresis,the authenticity of earthworm was identified,and the stability of the method was verified by experiments.Results it was found that the PCR products of the preparations containing earthworm had a single band between 260bp,and the conditions for identification were as follows:pre denaturation at 95℃for 5min,35 cycles(denaturation at 95℃for 30s,58℃for 30s,72℃for 30s),and extension at 72℃for 5min.
