摘　　要：目的蕲蛇是蝰科动物五步蛇Agkistrodon acutus(Giienther)的干燥体,具有祛风,通络,止痉的功效.近年来,由于资源有限,价格昂贵,出现了用其他蛇冒充蕲蛇或在蕲蛇体内掺假的现象.可采用分子生物学方法来鉴定蕲蛇的真伪,并建立一种鉴定蕲蛇的方法.方法制备蕲蛇水提物、醇提物.提取蕲蛇粉末、蕲蛇水提物、蕲蛇醇提物、市售蕲蛇以及风湿祛痛胶囊的DNA,采用PCR方法得到PCR产物,并电泳鉴定其产物大小,鉴定蕲蛇真伪,通过实验验证方法的稳定性.结果研究发现含有蕲蛇的制剂的PCR产物均在240 bp之间有单一条带,并摸索得出鉴定的条件为:95℃预变性5 min,35个循环(95℃变性30 s,64℃30 s,72℃30 s),72℃延伸5 min,经过验证具有稳定性.Agkistrodon acutus is the dried body of Agkistrodon acutus(gienther),an adder.It has the functions of expelling wind,dredging collaterals and stopping spasm.In recent years,due to the limited resources and high price,there has been a phenomenon of using other snakes to pretend to be Agkistrodon or adulterate in the body of Agkistrodon,so the molecular biological method is used to identify the authenticity of Agkistrodon,and a method is established to identify Agkistrodon.Methods water extract and alcohol extract of Agkistrodon acutus were prepared.The DNA of Agkistrodon acutus powder,Agkistrodon acutus water extract,Agkistrodon acutus alcohol extract,Agkistrodon acutus sold on the market and Fengshi Qutong capsule was extracted.The PCR product was obtained by PCR method,and its size was identified by electrophoresis,and the authenticity of Agkistrodon acutus was identified.The stability of the method was verified by experiments.Results the PCR products containing Agkistrodon acutus were found to have a single band between 240 BP.The conditions for identification were as follows:pre denaturation at 95℃for 5 min,35 cycles(denaturation at 95℃for 30 s,64℃for 30 s,72℃for 30 s),and extension at 72℃for 5 min.
