摘　　要：目的优选蔷薇红景天中总黄酮的提取及纯化工艺。方法采用Box-Behnken响应面法,以提取物中总黄酮含量为指标,考察提取溶剂体积分数、提取时间、提取次数、物料比等因素对提取工艺的影响;以总黄酮纯度为指标,考察样品溶液质量浓度、进样流速、洗脱液浓度、洗脱液体积等因素对纯化效果的影响。结果最佳提取工艺为70%乙醇,提取60 min,物料比1∶40;蔷薇红景天总黄酮提取物中的黄酮平均含量为56.97%;最佳纯化工艺为上样液质量浓度2.0035 mg/m L、洗脱液体积分数90%、洗脱液体积2 BV;纯化后,总黄酮平均纯度为82.83%,总黄酮含量比提取物的总黄酮含量提高了26%。结论实测值与理论预测值相符性良好,响应面法优化得到的蔷薇红景天总黄酮提取及纯化工艺参数条件可靠,具有一定的实际应用价值。Objective To optimize the extraction and purification of total flavonoids from Rhodiola rosea.Methods Box-Behnken response surface methodology was used to investigate the effects of solvent concentration;taking the total flavonoids content in the extract as an index,the extraction time,extraction times and material ratio on the extraction process were investigated;with purity of total flavonoids as an index,the effects of sample concentration,flow rate,eluent concentration and eluent volume on purification efficiency were investigated.Results The optimal extraction process was 70% ethanol concentration,60 min extraction time and 1∶40 material ratio.Under these conditions,the content of total flavonoids from Rhodiola rosea is 56.97%;the best purification process was 2.0035 mg/m L of the sample solution,90% of the eluent and 2 BV of the eluent.The purity of total flavonoids after purification was 82.83%.After purification,the content of total flavonoids was 26%higher than that of extract.Conclusion The measured values are in good agreement with the theoretical predictions.The optimal extraction and purification parameters of total flavonoids from Rhodiola rosea by response surface methodology are reliable and have certain practical application value.
