摘 要:目的探讨七叶亭通过调控Th1/Th2失衡对过敏性哮喘模型小鼠的抗哮喘作用。方法采用卵白蛋白(OVA)联合氢氧化铝凝胶多次激发致敏法造模。造模成功后第16~29天(连续14 d)七叶亭低、中、高剂量组小鼠腹腔注射七叶亭(100,200,400 mg/kg),地塞米松组小鼠腹腔注射地塞米松(1.25 mg/kg),对照组和模型组小鼠腹腔注射生理盐水。末次给药24 h后测定小鼠气道反应性,处死小鼠,进行肺组织病理学检查,测定肺泡灌洗液中干扰素γ(IFN-γ)、白细胞介素4(IL-4)和免疫球蛋白E(IgE)含量,以及脾脏中Th1和Th2细胞。结果地塞米松和不同剂量七叶亭干预均可降低气道阻力;对照组肺组织炎性细胞较少;模型组肺组织中炎性细胞明显浸润支气管周和血管外周结缔组织;给予地塞米松和七叶亭后,炎性细胞浸润明显减弱;与对照组比较,模型组,地塞米松组和七叶亭低、中、高剂量组小鼠IFN-γ含量和CD4^+IL-4^+细胞百分率升高,IL-4和IgE含量及CD4^+IFN-γ^+细胞百分率和Th1/Th2降低(P<0.05);与模型组相比,地塞米松组和七叶亭低、中、高剂量组小鼠IFN-γ含量和CD4^+IL-4^+细胞百分率降低,IL-4和IgE含量及CD4^+IFN-γ^+细胞百分率和Th1/Th2升高(P<0.05);七叶亭治疗效果呈剂量反应关系(P<0.05);地塞米松组和七叶亭高剂量组作用相似(P>0.05)。结论七叶亭通过调控Th1/Th2失衡可有效改善小鼠过敏性哮喘的进展,可能有抗过敏性哮喘的作用。Objective To investigate the anti-asthmatic effect of esculetin on allergic asthma model mice by regulating Th1/Th2 imbalance.Methods The ovalbumin(OVA)combined with aluminium hydroxide gel was used to induce allergic asthma model.From the16 th day to 29 th day(continuous for 14 d)after successful model establishment,the low,middle and high dose esculetin groups were intraperitoneally injected with esculetin(100,200,400 mg/kg),the dexamethasone group was intraperitoneally injected with dexamethasone(1.25 mg/kg)as positive control drug,and the control group and model group were intraperitoneally injected with saline.The airway responsiveness of mice was measured 24 h after the last administration.The mice were executed,and the lung histopathology was examined.The contents of interferon-γ(IFN-γ),interleukin-4(IL-4)and immunoglobulin E(IgE)in alveolar lavage fluid and Th1 and Th2 cells in spleen were determined.Results Intervention with dexamethasone and different concentrations of esculetin could reduce airway resistance;the control group had fewer inflammatory cells in lung tissue;the model group had obvious infiltration of inflammatory cells into peribronchial and perivascular connective tissue;after treatment with dexamethasone and esculetin,the infiltration of inflammatory cells were significantly reduced;compared with the control group,the content of IFN-γ and the percentage of CD4^+IL-4^+ cells in the model group,dexamethasone group and each esculetin group increased,the content of IL-4 and IgE,the percentage of CD4^+IFN-γ^+ cells and the percentage of Th1/Th2 were significantly decreased(P<0.05).After treatment with dexamethasone and esculetin,the content of IFN-γ and the percentage of CD4^+IL-4^+ cells were significantly decreased,the content of IL-4 and IgE and CD4^+IFN-γ^+ cells,Th1/Th2 ratio were significantly increased compared with model group(P<0.05);the therapeutic effect of each esculetin showed a dose-response relationship(P<0.05);the effects of dexamethasone group and high-dose esculetin gro
