摘  要:目的探讨天丹通络胶囊对急性脑梗死大鼠神经功能缺损评分及海马区勿动蛋白-A(Nogo-A)及溶血磷脂酸(LPA)蛋白表达的影响。方法将80只SD大鼠随机分为对照组、模型组、天丹通络胶囊低剂量组(40.0 mg/kg)、高剂量组(80.0 mg/kg),每组20只,雌雄各半。模型组、天丹通络胶囊各剂量组建立急性脑梗死模型。天丹通络胶囊各剂量组于造模成功第1天开始给予相应剂量药物灌胃,持续给予3个月,对照组和模型组给予等体积生理盐水。评价各组大鼠神经运动功能,观察大鼠脑梗死区域病理改变,采用反转录-聚合酶链反应(RT-PCR)法及蛋白质印迹(Western Blot)法测定脑梗死海马组织Nogo-A及LPA的mRNA和蛋白表达水平。结果治疗7,14 d后,模型组神经功能缺损评分、双侧贴纸去除时间、平衡木过杆时间、海马Nogo-A及LPA的mRNA和蛋白表达水平明显高于对照组(P<0.05);天丹通络胶囊低、高剂量组上述指标均明显低于模型组(P<0.05),且随着天丹通络胶囊给药剂量的增加而逐渐降低(P<0.05);对照组海马组织神经细胞正常;模型组海马组织见大量坏死神经元,呈缺血样病理改变,空泡样变性坏死数量较多;天丹通络胶囊高剂量组海马组织神经细胞坏死有明显改善;天丹通络胶囊低剂量组神经元疏松紊乱,但坏死神经元细胞减少。结论天丹通络胶囊对急性脑梗死大鼠神经功能缺损有防护作用,其作用机制可能与其通过抑制脑梗死海马区Nogo-A及LPA的mRNA和蛋白表达,进而抑制神经元细胞的坏死与凋亡有关。Objective To investigate the effect of Tiandan Tongluo Capsules on neurological deficit score and Nogo-A,lysobisphosphatidic acids(LPA)protein expression in hippocampus of rats with acute cerebral infarction.Methods Eighty SPF grade SD rats were randomly divided into four groups:control group,model group,Tiandan Tongluo Capsules low dose group(40.0 mg/kg),Tiandan Tongluo Capsules high dose group(80.0 mg/kg),20 rats in each group,half male and half female.Acute cerebral infarction model was established in the model group and the two doses of Tiandan Tongluo Capsules groups.Each dose group of Tiandan Tongluo Capsules was given the corresponding dose of medicine by gavage on the first day after the successful model establishment,which lasted for 3 months.The rats in the control group and the model group were given the same volume of normal saline.At the end of the experiment,the neuromotor function,pathological changes of cerebral infarction area were examined,and the Nogo-A,LPA gene and protein levels in hippocampus were evaluated by reverse transcription polymerase chain raction(RT-PCR)and Western Blot.Results The neurological deficit scores at 7,14 d after treatment,the removal time of bilateral stickers,the passing time of the balance beam,the expression level of Nogo-A,LPA mRNA and protein in hippocampus in the model group were significantly higher than those in the control group(P<0.05).The low dose group and high dose group of Tiandan Tongluo Capsules were significantly lower than those in the model group(P<0.05).With the increase of the dosage of Tiandan Tongluo Capsules,the above indicators were gradually decreased(P<0.05).In the control group,the neurons were normal.In the model group,a large number of necrotic neurons were observed,which showed a lack of blood like pathological changes and a large number of vacuolated degeneration necrosis.In the high dose group of Tiandan Tongluo Capsules,the necrotic neurons were significantly improved.In the low dose group of Tiandan Tongluo Capsules,the neurons were l
