摘  要:以红花绿绒蒿为试材,采用正交实验设计,研究了酶切时间、Mg^2+用量、dNTPS用量、模板DNA稀释倍数等对AFLP反应体系的影响,以期建立一个适合红花绿绒蒿的AFLP反应体系,并用优化后的体系对64对AFLP引物进行筛选。结果表明:通过所建立的AFLP反应体系能够获得清晰的条带,并利用该体系筛选出8对适合于红花绿绒蒿AFLP分析的引物组合。该研究可为今后AFLP标记在绿绒蒿属植物的遗传多样性分析及种质鉴定提供参考依据。The Meconopsis puniceawas used as template in this study.Used orthogonal experimental design,the effect of relevant factors such as the digestion time,the amount of Mg^2+,the amount of dNTPS,and the template DNA dilution factor on the AFLP reaction system were studied.The goal was to establish an AFLP reaction system suitable for M.puniceaand screen 64 pairs of AFLP primers with the optimized system.The results showed that the clear bands could be obtained through the established AFLP reaction system,and 8 pairs of primer combinations suitable for AFLP analysis of M.punicea were screened by using this system.This study provided a certain reference for future genetic diversity analysis and germplasm identification of AFLP markers in M.Vig.
