摘 要:目的探讨不同实验剂量附子乌头碱对正常615小鼠外周血单核细胞分泌前列腺素E2(PGE2)的表达影响及其对615小鼠外周单核细胞分化成调节T细胞(Treg)的调节作用。方法分离健康小鼠外周血单核细胞,分别以不同实验剂量附子乌头碱高、中、低剂量(2、0.5、0.1 mg/mL)作为实验组及塞来昔布为阳性对照处理24 h,然后测定其受到不同剂量附子乌头碱及塞来昔布刺激后分泌PGE2和分化CD4+CD25+Foxp3+Treg的水平。采用ELISA检测PGE2的浓度,流式细胞术检测CD4+CD25+Foxp3+Treg细胞比例。结果与阴性对照组比较,乌头碱高剂量能显著降低PGE2含量且有显著性差异(P<0.05),而PGE2分泌量随乌头碱剂量的增加而降低;与阴性对照组比较,乌头碱可降低Treg分化比例,乌头碱高、低剂量组有极显著性差异(P<0.01),乌头碱中剂量组有显著性差异(P<0.05),乌头碱低剂量组Treg比例反而增加,而Treg比例随乌头碱剂量的增加而减少。可见高、中剂量附子乌头碱能导致小鼠外周血单核细胞分泌PGE2减少,同时抑制了CD4+CD25+Foxp3+Treg的分化。结论适当剂量(高、中剂量)附子乌头碱可减弱615小鼠外周血单核细胞产生免疫抑制。附子乌头碱抗免疫抑制作用在实验所选剂量范围中呈相关性,且对抗免疫抑制具有双向调节作用。Objective To investigate the effects of aconitine on the secretion of prostaglandin E2(PGE2)by peripheral blood mononuclear cells of normal 615 mice and its regulation on the differentiation of peripheral mononuclear cells into regulatory cell(Treg)in 615 mice.Methods Monocytes from peripheral blood of healthy mice were isolated and treated with different doses of aconitine(2,0.5,0.1 mg/mL)as experimental group and celecoxib as positive control for 24 hours.The levels of PGE2 secretion and CD4+CD25+Foxp3+Treg differentiation were determined after stimulation with different doses of aconitine and celecoxib.The concentration of PGE2 was detected by ELISA and the proportion of CD4+CD25+Foxp3+Treg cells was detected by flow cytometry.Results Compared with negative control group,aconitine at high dosage could significantly reduce the content of PGE2(P<0.05),while the secretion of PGE2 decreased with the increase of aconitine dosage;compared with negative control group,aconitine could reduce the proportion of Treg differentiation,aconitine at high and low dosage group had significant difference(P<0.01),aconitine at medium dosage group had significant difference(P<0.05).Treg ratio increased in low dose group,but decreased with the increase of aconitine dose.It could be seen that aconitine in high and medium doses can reduce the secretion of PGE2 by peripheral blood monocytes and inhibit the differentiation of CD4+CD25+Foxp3+Treg.Conclusion Appropriate doses(high and medium doses)of aconitine can attenuate immunosuppression of peripheral blood mononuclear cells in 615 mice.The antiimmunosuppressive effect of aconitine is correlated with the dose range selected in the experiment,and it has two-way regulating effect on anti-immunosuppressive effect.
