摘  要:目的探讨平喘宁对支气管哮喘模型大鼠气道黏液高分泌的调控机制。方法 SPF级别雄性SD大鼠140只,随机分为正常组,模型组,平喘宁低、中、高剂量组、桂龙咳喘宁组及地塞米松组,每组20只。以卵清蛋白(OVA)联合氢氧化铝复制大鼠哮喘模型,造模28 d后,正常组、模型组每天给予等量蒸馏水灌服,平喘宁低、中、高剂量组按0.49、0.98、1.96 g/(kg·d)灌胃给药,桂龙咳喘宁按0.405 mg/(kg·d)灌胃给药,地塞米松组按0.5 mg/(kg·d)灌胃给药。灌胃4周后,在末次OVA激发后2 h内解剖大鼠并取出肺组织。采用HE染色法观察肺组织的病理形态学改变;采用RT-PCR法检测肺组织MUC5AC mRNA的表达;采用免疫荧光法与Wersten-blot法检测肺组织中MUC5AC蛋白表达量;ELISA法检测支气管肺泡灌洗液(BALF)中MUC5AC的分泌水平。结果与正常组比较,模型组肺组织病理切片观察可见炎性细胞浸润,气道壁增厚,气道狭窄、变形,肺泡增大、断裂,间隔增厚及气道平滑肌增厚;MUC5AC阳性表达IOD值明显增加(P<0.01);MUC5AC蛋白及mRNA表达水平升高(P<0.01)。与模型组比较,各治疗组大鼠肺组织病理形态学结构改善;MUC5AC阳性表达IOD值降低(P<0.01);MUC5AC蛋白及mRNA表达水平下降(P<0.01)。结论平喘宁对支气管哮喘具有治疗效果,作用机制可能与调节MUC5AC水平、减少气道黏液高分泌有关。Objective To observe the effect of Pingchuanning on the model rats of bronchial asthma,to analyze the regulation and control level of the mucin AC(MUC5 AC)in the bronchial asthma rats,and to explore the mechanism of relieving asthma,warming the lung,reducing phlegm,relieving cough and asthma. Methods 140 male SD rats were randomly divided into normal group,model group,Pingchuanning low,middle,high dose group,Guilong Kechuanning group and dexamethasone group,20 rats each group.The asthma model of the rat was duplicated with ovalbumin(OVA)and aluminum hydroxide. After the model was established for 28 d,the normal group and the model group were given equal amount of distilled water daily,Pingchuanning low,middle and high dose groups were given 0.49,0.98,1.96 g/(kg·d)intragastrically,Guilong Kechuanning was given 0.405 mg/(kg·d)intragastrically,the dexamethasone group was given 0.5 mg/(kg·d)intragastrically. After 4 weeks,the rats were dissected within 2 hours after the last ovalbumin challenge and the lung tissue was taken out. Hematoxylin-eosin(HE)staining was used to observe the pathomorphological changes of lung tissue,RT-PCR method was used to detect the expression abundance of MUC5 AC in lung tissue,the relative expression of MUC5 AC in lung tissue was detected by immunofluorescence and Wersten-blot,and the level of MUC5 AC in bronchoalveolar lavage fluid(BALF)was detected by ELISA. Results Compared with the normal group,inflammatory cell infiltration,thickening of airway wall,airway stenosis,deformation,alveolar enlargement,rupture,septum thickening and airway smooth muscle thickening were observed in the model group,the positive expression of IOD in the model group was significantly higher than that in the normal group,the expression of MUC5 AC protein and m RNA in the model group was significantly higher than that in the normal group(P<0.01). Compared with the model group,the pathomorphological structure of lung tissue in each treatment group was improved. The positive expression of IOD in MUC5 AC in each
