HPLC法测定肝豆灵片的盐酸小檗碱和芦荟大黄素含量

摘　　要：目的 :采用HPLC法测定肝豆灵片中盐酸小檗碱、芦荟大黄素的含量,建立肝豆灵片中指标性成分的含量测定分析方法。方法 :色谱条件:C 18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈(B):0.03M磷酸二氢钠-0.1%磷酸(A),等度洗脱(B∶A=38∶62),λmax=225 nm,流速1.0 mL/min,柱温30℃,分析时间30 min。结果 :混合标准溶液中盐酸小檗碱浓度1.25μg/mL^80μg/mL、芦荟大黄素浓度0.037 5μg/mL^2.4μg/mL,线性范围良好(r盐酸小檗碱=0.999 4,r芦荟大黄素=0.998 9),精密度RSD分别为1.42%、1.57%;稳定性RSD分别为1.84%、1.86%;重复性RSD分别为1.28%、1.37%;加样回收率分别为99.16%、99.73%;平均加样回收率RSD分别为1.41%、1.30%。结论 :实验结果表明,该方法测定肝豆灵片中盐酸小檗碱、芦荟大黄素的含量,灵敏度高,操作简便,测定结果准确可靠,可为肝豆灵的质量控制提供理论支持。Objective:To determine the contents of berberine hydrochloride and aloe-emodin in Gandouling tablet by HPLC,and the application value of HPLC in the quality control of Gandouling production.Method:Chromatographic conditions:C18 chromatographic column(4.6 mm×250 mm,5μm),mobile phase:methyl cyanide(B):0.03M primary sodium phosphate^0.1%phosphoric acid(A),isocratic elution(B:A=38:62),λmax=225nm,flowrate 1.0 mL/min,column temperature 30℃,analysis time 30 min.Result:The concentration of berberine hydrochloride in the mixed standard solution was 1.25μg/mL^80μg/mL、The concentration of aloe emodin was 0.0375μg/mL^2.4μg/mL.The linear range was good(r1=0.9994,r2=0.9989).The RSD of the precision is 1.42%and 1.57%.The RSD of the stability is 1.42%and 1.57%,The RSD of the repeatability is 1.28%and 1.37%.The RSD of the returning rate is 99.16%and 99.73%.The RSD of the average recovery is 1.41%and 1.30%.Conclusion:The experimental results showed that the method was sensitive,simple and reliable for the determination of berberine hydrochloride and aloe emodin in Gandouling medicine,providing scientific method for the quality control of Gandouling.
