摘 要:目的:探讨当归四逆汤对糖尿病大鼠坐骨神经miR-210、HIF-1α和VEGF表达的影响。方法:30只SD大鼠分为空白对照组和观察组,观察组采用1%链脲佐菌素50 mg/kg诱导生成糖尿病大鼠模型,造模成功的观察组大鼠17只按随机数字表法分为糖尿病模型组和中药预防组。MP组灌服当归四逆汤(12 mL/kg/d),其余组灌服生理盐水,12周结束后,检测3组大鼠空腹血糖、光热刺激痛阈时间及坐骨神经传导速度,并取坐骨神经检测miR-210、HIF-1αmRNA水平及VEGF表达。结果:12周后与BC组比较,DM组空腹血糖、光热刺激痛阈时间明显升高,坐骨神经传导速度、miR-210、HIF-1αmRNA及VEGF表达明显降低,差异有统计学意义。与DM组比较,MP组大鼠光热刺激痛阈时间明显降低,坐骨神经传导速度、miR-210、HIF-1αmRNA及VEGF表达明显增高,差异有统计学意义。结论:早期预防性应用当归四逆汤可改善糖尿病周围神经病变,其机制可能与上调神经组织miR-210、HIF-1αmRNA及VEGF表达有关。Objective:To observe the effects of the Chinese Herbal Medicine Prescription Danggui Sini Decoction for Frigid Extremities(DDFE)on miR-210,HIF-1αand VEGF expression of Sciatic Neuropathy in Streptozotocin-Induced Diabetic Rats.Methods:30 SD rats were randomly assigned to two groups:the observation group(n=22)and blank control group(BC group,n=8).The observation group received a single intraperitoneal injection of 1%streptozotocin(50mg/kg body weight)to prepare the model group of Diabetic Rats.17 rats after successful modeling in the observation group were randomly divided into diabetic model group(DM group,n=8)and medicine prophylaxis group(MP group,n=9).The DDFE at a dosage of 12 mL/kg/d was administered to the rats in the MP group,while the other rats were given distilled water at the same dosage.Fasting blood glucoses,thermal thresholds,sciatic nerve conduction velocities,and the levels of miR-210,HIF-1 mRNA and VEGF protein in sciatic nerves were measured after 12 weeks.Results:Fasting blood glucoses,thermal thresholds were significantly increased while sciatic nerve conduction velocities,levels of miR-210,HIF-1 mRNA and VEGF protein were significantly decreased compared with BC group 12 weeks later.Thermal thresholds were significantly decreased while sciatic nerve conduction velocities,levels of miR-210,HIF-1 mRNA and VEGF protein were significantly increased compared with DM group after 12 weeks of treatment with DDFE.Conclusion:Early preventive application of DDFE may delay the development of DPN via increasing miR-210,HIF-1αmRNA and VEGF expression in the sciatic nerves of diabetic rats.
