补肾抗衰片对动脉粥样硬化模型血管周围脂肪炎症的实验研究

摘　　要：[目的]研究补肾抗衰片对动脉粥样硬化模型血管周围脂肪组织(PVAT)炎症的干预作用。[方法]选取雄性新西兰兔24只,将其随机分为对照组、模型组、补肾抗衰组、阿托伐他汀钙组,以高脂饲料喂养法建立动脉粥样硬化模型。高脂喂养4周后,两给药组分别给予补肾抗衰片、阿托伐他汀钙片,连续干预8周后,取附着血管外周脂肪的主动脉行苏木精-伊红(HE)染色,观察主动脉及PVAT组织病理形态学变化,采用免疫组化法检测主动脉及PVAT组织瘦素、肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-9(MMP-9)蛋白的表达。[结果]补肾抗衰片可以减轻内膜增生程度,使中内膜厚度比下降;减少PVAT组织巨噬细胞浸润,减小脂肪细胞直径;使主动脉和PVAT组织TNF-α、瘦素、MMP-9阳性颗粒表达面积减小。[结论]补肾抗衰片对动脉粥样硬化模型PVAT炎症的表达具有抑制作用。[Objective]The research aimed at exploring the effect of BushenKangshuai tablet on perivascular adipose tissue(PVAT)inflammation of atherosclerosis model rabbit.[Methods]Male New Zealand rabbits were randomly divided into normal control group,model group,Bushen Kangshuai Tablets group and atorvastatin calcium group.High fat diet was used to replicate the atherosclerosis model.After 4 weeks of high-fat feeding,Bushen Kangshuai Tablets was given to the Bushen Kangshuai Tablets group,atorvastatin calcium was given to the atorvastatin group.After 8 weeks of continuous intervention,samples were collected.Retain the thoracic aorta with PVAT,one part using HE staining to observe the histopathological changes of aorta and PVAT,another using immunohistochemical method to detect the expression of the aorta and PVAT organization leptin,TNF-α,MMP-9 protein.[Results]Bushen Kangshuai Tablet could reduce the degree of intimal hyperplasia and decrease the ratio of intimal thickness.Reduce the infiltration of macrophages in PVAT tissue and reduce the diameter of fat cells.The expression areas of TNF-α,leptin and MMP-9 positive particles in aorta and PVAT were decreased.[Conclusion]Bushen Kangshuai Tablets has inhibitory effect on theexpression of perivascular adipose tissue inflammation in atherosclerosis model.
