正交试验法优化芪芍醒脑颗粒醇提工艺

摘  要:研究芪芍醒脑颗粒的最佳醇提工艺,以含芍药苷的量为评价指标,采用正交试验法设计优化最佳提取工艺.芍药苷含量测定色谱条件:Diamonsil C18色谱柱(4. 6 mm×250 mm,5μm);流动相为乙腈-0. 1%磷酸溶液(14∶86);流速为1. 0 m L/min;柱温23℃.结果表明,芪芍醒脑颗粒的最佳醇提工艺为6倍量70%乙醇,加热回流提取3次,每次2 h.优选的醇提工艺合理、可行,有效成分高,为芪芍醒脑颗粒提取工艺的确定提供了试验依据.Study on the optimum ethanol extraction technology of Qishao Xingnao granules,taking the content of paeoniflorin as the comprehensive evaluation index,theoptimum extraction process was optimized by orthogonalexperimentdesign.Chromatographic conditions for the determination of paeoniflorin Diamonsil C 18 chromatographic column(4.6 mm×250 mm,5μm);the mobile phase was acetonitrile-0.1%phosphoric acid solution;current speed:1.0 mL/min;column temperature:23℃.The results showed that the optimum ethanol extraction process of Qishao Xingnao granules was determined as 6 times of 70%ethanol,heated and refluxed for 3 times,2 hours each time.The optimized alcohol extraction process was reasonable and feasible,with high content of active components,which provided experimental basis for the determination of the extraction process of Qishao Xingnao granules.
