摘  要:目的:观察滨蒿总黄酮体外抗流感病毒作用及对TLR3/NF-κB信号通路的影响。方法:Cytopathic effect (CPE)法观察滨蒿黄总酮0. 125 mg/ml、0. 25 mg/ml体外抗流感病毒的作用;TRIzol法提取总RNA,用RT-qPCR法测定TLR3/NF-κB信号通路靶点和炎症因子的mRNA水平。结果:滨蒿总黄酮在体外对流感病毒甲型、乙型均有明显的抑制作用,随着浓度的增加,抗病毒作用增强,呈量效关系,TI(药物治疗指数)值约为15;进一步实验发现与病毒对照相比,滨蒿总黄酮0. 25 mg/ml可明显下调TLR3/NF-κB信号通路靶点TLR3、NF-κB、IRF3、IRF7 mRNA表达水平,对流感病毒诱导表达的炎症因子IL-6、MCP-1、i NOS、Cox2有明显的干预作用(P <0. 05或P <0. 01)。结论:滨蒿总黄酮通过抑制TLR3/NF-κB信号通路发挥免疫调节作用,从而达到抗流感的效应。Objective: To investigate the anti-influenza effects and mechanism of total flavonoids from Artemisia scoparia Waldst. et kit.( ASTF)on TLR3/NF-κB signaling pathway in vitro. Methods: Cytopathic effect( CPE) was used to evaluate the anti-influenza virus effect of ASTF( 0. 125 mg/ml、0. 25 mg/ml) in vitro. Total RNA was extracted by TRIzol method,the TLR3/NF-κB signaling pathway and mRNA level of inflammatory factors were determined by RT-q PCR. Results: ASTF showed the inhibitory effect on influenza A and B virus in vitro. As the concentration increased,the antiviral effect was enhanced,in a dose-dependent way. The therapeutic index( TI) value was approximately15. Further experiments found that 0. 25 mg/m L ASTF reduced the mRNA expression levels of TLR3,NF-κB,IRF3,IRF7,and significantly reduced the mRNA expression levels of IL-6,MCP-1,i NOS,Cox2 induced by influenza virus. Conclusion: The anti-influenza effect of ASTF plays an immunomodulatory effect by inhibiting the targets on TLR3/NF-κB signaling pathway,thus achieving the anti-influenza effect.
