摘 要:目的:采用注射自体血栓的方法制备大鼠局灶脑梗塞模型,观察舒脑欣滴丸对脑中风恢复期治疗作用。方法:实验设假手术对照组、模型对照组、舒脑欣滴丸13、26、52、104 mg/kg剂量组和阳性对照药脑心通500 mg/kg组,每组10只。脑缺血后第3 d开始给药,每天1次,连续给药30天。通过平衡木测验观察大鼠肢体功能;通过前肢抓力测定,观察肌力功能;通过避暗穿梭试验,观察大鼠学习记忆功能;通过脑组织HE染色,显微镜病理学,观察海马、皮层神经元数目以及病理学变化;通过免疫组化法,检测GFAP表达。结果:与模型对照组比较,舒脑欣滴丸能显著增加大鼠运动功能评分及前肢抓力;明显降低增加的受电击次数;明显增加海马CA1区和皮层神经元数目,并增加海马CA1区GFAP的表达(P <0. 05或P <0. 01)。结论:舒脑欣滴丸对大鼠脑中风恢复期有治疗作用,改善其神经元功能,增强学习记忆,可能与GFAP表达增加进而促进再生有关。Objective: To explore the protective effect of Shunaoxin dropping pill in rats in the recovery stage of cerebral stroke induced by autologous thrombus. Methods: Rats were divided into the sham operation group,the model group,Shunaoxin groups( 13 mg/kg,26 mg/kg,52 mg/kg,104 mg/kg) and the positive control group( Naoxintong 500 mg/kg),10 rats in each group. 3 days after cerebral ischemia was established,corresponding drugs were administered daily for 30 consecutive days. The limb function was observed by balance wood test,the muscular tension was observed by forelimb grasping force measurement. The ability of learning and memory was observed by defensive shuttle test. Pathological changes and numbers of neurons in hippocampus and cortex were observed by HE staining and microscope pathology. GFAP expression was determined by immunohistochemical method. Results : Compared with the model group,Shunaoxin dropping pill improved the impaired motor function and forelimb grasping force,reduced the increasing numbers of electric shocks and increased the numbers of neurons in hippocampus CA1 and cortex,also enhanced the expression of GFAP in hippocampus CA1. Conclusion: Shunaoxin dropping pill has the therapeutic effect in rats in the recovery stage of stroke,it improves neuronal function of learning and memory,which may be related to increasing of GFAP expression and promoting its regeneration.
