摘 要:【目的】探究家蚕感染BmNPV后,不同组织碱性磷酸酶(ALP)及其基因表达变化规律,为阐明BmNPV的侵染机制及培育抗NPV蚕品种的选育提供借鉴和思考。【方法】通过经口添食BmNPV,检测分析家蚕中肠、血淋巴和脂肪体ALP基因的表达水平及其编码的碱性磷酸酶活性变化情况。【结果】家蚕中肠ALP基因在感染BmNPV后6、9和12 h呈现上调表达趋势,其编码的碱性磷酸酶活性显著高于对照组,在感染24 h基因表达被抑制,同时酶活性显著降低。血淋巴碱性磷酸酶及ALP基因对BmNPV的响应略晚于中肠,ALP基因在感染9 h开始上调表达,在12 h表达量最高,在24 h表达量急剧降低。碱性磷酸酶活性在9和12 h极显著高于对照组,而在24 h急剧减弱,显著低于对照组。脂肪体ALP基因表达量及碱性磷酸酶活性仅在感染12 h显著高于对照组。【结论】家蚕感染BmNPV后,中肠、血淋巴和脂肪体ALP基因表达水平及碱性磷酸酶活性变化均是呈现先升高后急剧减弱的趋势。基因的表达情况与酶活性变化规律一致,这与家蚕感染BmNPV后,其自身机体的生理代谢进程存在紧密联系,暗示碱性磷酸酶在家蚕抗病毒过程中发挥了重要作用。【Objective】In order to provide reference and consideration to clarify the infection mechanism of BmNPV and breed varieties resistant to BmNPV,the present study explored the changes of alkaline phosphatase(ALP)activity and its gene expression in different tissues of Bombyx mori after infection with BmNPV.【Method】The real-time fluorescent quantitative PCR was applied to analyze the expression of ALP,meanwhile the activities of the alkaline phosphatase were determined in midgut,haemolymph and fat body of silkworm larvae after feeding BmNPV.【Result】In midgut of silkworm,the ALP expression was up-regulated at 6,9 and 12 hours after infection with BmNPV,meanwhile the alkaline phosphatase activity was significantly higher than that in the control group.Whereas the ALP activity was significantly reduced and its gene was inhibited at 24 hours.The response to BmNPV was slightly later in haemolymph than that in the midgut,the expression of ALP was up-regulated at 9 hours and reached the highest level at 12 hours,however it was decreased sharply at 24 hours after infection.The alkaline phosphatase activity was significantly higher at 9 and 12 hours,but significantly lower at 24 hours than that in the control group.In fat body,the alkalinephosphatase activity and its gene expression were significantly higher than that in the control group only at 12 hours after infection with BmNPV.【Conclusion】The expression levels of ALP and alkaline phosphatase activity were all increased first and then decreased sharply in the midgut,heamolymph and fat body of silkworm after infection with BmNPV.The gene expression were basically consistent with the change of enzyme activity,which was closely related to the physiological metabolic process of silkworm after infection with BmNPV.It suggested that the alkaline phosphatase played an important role in the antiviral process of silkworm.
