摘  要:水杉(Metasequoia glyptostroboides)是著名的杉科孑遗植物,对水杉的分子遗传研究在其资源保护和利用方面具有重要意义。本研究以水杉为实验材料,在全长转录组测序的基础上克隆了一个泛素结合酶基因(MgUC-E2),并进一步对该基因作了生物信息学分析。结果表明:MgUC-E2基因cDNA编码链全长为691 nt,可以编码长度为148个氨基酸的泛素结合酶。该酶蛋白的分子量为16545.09 kD,等电点为7.72,平均吸水值为-0.343;含有15个磷酸化修饰位点和5个常见功能位点;在预测的二级结构中共包括5个螺旋区、4个折叠区和一些无规则卷曲区;三级结构分析的结果进一步证实MgUC-E2基因就是编码泛素结合酶。本研究结果可为水杉及其它相关物种的泛素结合酶基因的结构及功能研究提供一些参考。Metasequoia glyptostroboides is a famous relict plant belonging to Taxaceae,it is great significant to study Metasequoia glyptostroboides at molecular genetics level for its conservation and utilization.In this study,Metasequoia glyptostroboides was as experimental material,a ubiquitin-conjugating enzyme gene(MgUC-E2)was cloned,and bioinformatic analysis.The results demonstrated that its cDNA coding strand was 691 nt in length,encoding ubiquitin-conjugating enzyme with 148 amino acids.The molecular weight of the protein was 16545.09 kD,the isoelectric point was 7.72,grand average hydropathicity value was-0.343;there were 15 phosphorylation modification sites and 5 common functional sites;its secondary structure included 5 helical regions,4 folding regions and some coil regions;tertiary structure analysis result further revealed that MgUC-E2 gene cloned here could encode ubiquitin-conjugating enzyme.The results of this study can provided some reference for the the studies on the structure and function of ubiquitin-conjugating enzyme gene in Metasequoia glyptostroboides and other related species.
