摘 要:本研究以转入甜菜碱醛脱氢酶基因(badh)的玉米自交系‘丹988’的T7代自交系为试验材料,利用断水及盐溶液分别模拟干旱胁迫与盐碱胁迫,对转基因材料进行苗期的抗旱性与耐盐碱性鉴定。鉴定结果表明,在抗旱性试验中,断水15 d、断水20 d时转基因玉米自交系各项抗旱指标数值普遍高于对照,且大部分存在显著性差异;在断水25 d时转基因玉米自交系与对照的全部抗旱指标存在显著性差异。在耐盐碱试验中,在胁迫20 d、30 d、40 d时丹988-BADH红-1、丹988-BADH红-2各项耐盐碱指标数值普遍高于对照,且大部分存在显著性差异;丹988-BADH红-4在胁迫20 d时部分耐盐碱指标低于对照,胁迫30 d、40 d时高于对照但未出现显著性差异。经隶属函数法抗性分析,得到转基因玉米自交系与对照的抗旱能力排序:丹988-BADH红-4>丹988-BADH红-2>丹988-BADH红-1>对照;耐盐碱能力排序:丹988-BADH红-1>丹988-BADH红-2>丹988-BADH红-4>对照。得出结论,转基因玉米自交系抗旱性与耐盐碱性全部优于对照,证明外源badh基因的导入提高了受体自交系的抗旱性与耐盐碱性,并得到了抗旱性、耐盐碱性较强且保持原有农艺性状的高世代转基因玉米自交系。In this study,the T7 inbred line of maize inbred line’Dan988’,which was transfected with betaine aldehyde dehydrogenase gene(badh),was used as the experimental material,and the drought resistance and saline-alkali tolerance of the transgenic material at seedling stage were identified by pot culture,simulated drought stress with cut-off water and simulated saline-alkali stress with salt solution.The results showed that in the drought resistance test,the indexes of the transgenic maize inbred lines were generally higher than those of the recipient inbred lines at 15 d and 20 d,and most of them had significant differences.There were significant differences in all indexes between the inbred lines and the receptors of transgenic maize at 25 d after water cut.In the salt-alkali tolerance test,the indexes of BADH red-1 and BADH red-1 were generally higher than those of the recipient inbred lines at the stress of 20 d,30 d and 40 d,and most of them were significantly different.Some indexes of BADH red-4 were lower than that of the receptor at 20 d of stress,and higher than that of the receptor unit at 30 d and 40 d of stress,but there was no significant difference.According to the resistance analysis by membership function method,the drought resistance sequence of transgenic maize inbred lines and recipient inbred lines was obtained:Dan 988-BADH red-4>Dan 988-BADH red-2>Dan 988-BADH red-1>CK;Sequence of salt and alkali tolerance:Dan 988-BADH red-1>Dan 988-BADH red-2>Dan 988-BADH red-4>CK.The above results indicated that the drought resistance and salt-alkali tolerance of transgenic maize inbred lines were all better than the control,which proved that the exogenous badh gene improved the drought resistance and salt-alkali tolerance of the recipient inbred lines,and the high-generation transgenic maize inbred lines with strong drought resistance,salt-alkali tolerance and maintained the original agronomic traits were obtained.
