摘 要:目的:观察针刺单穴与腧穴配伍干预骨肉瘤模型小鼠相关的形态学及免疫学效应差异。方法:构建骨肉瘤K7皮下荷瘤模型小鼠,随机分为空白组、模型组、单穴组和配穴组,单穴组造模后针刺单穴(肾俞),配穴组造模后针刺腧穴配伍(百会、肾俞、足三里),应用多模式光学活体成像技术(In-Vivo Xtreme技术)和游标卡尺观察小鼠肿瘤大小变化,利用酶联免疫吸附法观察血清细胞因子IL-6、IFN-γ和脾脏组织中TNF-α等相关免疫因子的变化情况,利用流式细胞仪检测相关免疫细胞如T细胞、NK细胞等免疫相关指标变化情况。结果:针刺治疗后,与模型组比较,各针刺组小鼠体质量、肿瘤大小显著降低(P<0.01),肿瘤信号强度显著降低(P<0.01);各针刺组NK细胞、细胞毒性T细胞含量显著下降(P<0.05,P<0.01),辅助性T细胞含量显著升高(P<0.05,P<0.01);各针刺组IL-6、TNF-α含量显著升高(P<0.01)。与单穴组比较,配穴组其各指标疗效显著改善(P<0.05,P<0.01)。结论:针刺治疗改善机体免疫相关指标,提高机体免疫力,可通过调节机体相关免疫因子达到干预肿瘤的效果,且配穴组疗效优于单穴组。Objective:To observe the difference in immunological and physiological effects between acupuncture at single acupoint and compatible acupoints in the treatment of osteosarcoma model mice.Methods:Osteosarcoma K7 subcutaneous tumor-burdened model mice were randomly divided into blank control group,model control group,single acupoint group and compatible acupoints group.The single acupoint group established model then received single point Shenshu(BL23),compatible acupoints group established model then received acupoints compatibility Baihui(DU20),Shenshu(BL23)and Zusanli(ST36).The multimode optical imaging in vivo and vernier caliper were used to observe the change of mice tumor size.Enzyme linked immunosorbent assay(ELISA)was used to observe the changes in immune-related indicators of the serum cytokine IL-6,IFN-γand spleen tissues TNF-αimmune factors.Flow cytometry was used to detect the changes of immune-related indicators,such as the immune-related indicators changes of T cells and NK cells.Results:After the acupuncture treatment,compared with the model control group,there were significant differences in body weight and tumor size among acupoints groups(P<0.01),and there were significant differences in tumor signal intensity(P<0.01).The content of NK cells,cytotoxic T cells among acupoints groups significantly decreased(P<0.05,P<0.01),and the content of helper T cells among acupoints groups significantly increased(P<0.05,P<0.01).The content of IL-6 and TNF-αamong acupoints groups significantly increased(P<0.01).Compared with the single acupoint group,other indicators in compatible acupoints group significantly improved(P<0.05,P<0.01).Conclusion:Acupuncture treatment can improve the immune-related indicators of the body,improve the immunity of the body,and achieve the effect of intervention in tumor by adjusting the immune-related factors of the body,and compatible acupoints group are better than that in single acupoint group.
