清热活血组分对急性脑梗死后患者miRNA/VEGF表达的影响

摘　　要：目的:研究清热活血组分对急性脑梗死后患者血清中miRNA/血管内皮生长因子(VEGF)表达的影响。方法:共纳入120例急性脑梗死受试者,应用中央随机系统随机分为4组:对照组、清热组、活血组、清热活血组。分别在第1、3、5、14天对受试者进行疗效性指标国立卫生研究院卒中量表(NIHSS)评分及血清中VEGF含量测定;同时,在第1、14天对患者血清中6种miRNA含量进行测定。结果:14d治疗之后,4组治疗均可改善NIHSS评分;与对照组同期比较,清热活血组在治疗第3天显著上调VEGF表达(P<0.05);清热活血组第5天VEGF含量差值较其他组显著升高(P<0.05);清热活血组在6种miRNA基因表达上均有升高作用,同时对hsa-miR-218-5p和hsa-miR-185-5p两种miRNA基因表达较其他组有显著的促进作用(P<0.05)。结论:清热活血组分可能通过促进对急性脑梗死后miRNA/VEGF的表达,改善患者NIHSS评分即神经功能缺损症状,从而发挥脑保护作用。Objective: To study the effects of heat-clearing and blood-activating components on the expression of miRNA/VEGF in serum of patients after acute cerebral infarction. Methods: A total of 120 subjects with acute cerebral infarction were included and randomly divided into 4 groups by central random system, namely the basic treatment group, heat-clearing group, blood-activating group and heat-clearing and blood-activating group. NIHSS score and serum VEGF content were measured on day 1, day 3, day 5 and day 14, respectively. Meanwhile, the contents of 6 mirnas in the serum of the patients were measured on day 1 and day 14. Results: On the 14 th day after treatment, NIHSS score of all four groups decreased. Compared with the basic treatment group, the effect of up-regulated VEGF expression on the 3 rd day of heat-clearing and blood-activating group was significant(P<0.05). The difference of VEGF content in the heat-clearing and blood-activating group on day 3 and 5 was significantly higher than that in the other groups(P<0.05). Compared with other groups, the heat-clearing and blood-activating components had significantly increased the expression of six mi RNA genes(P<0.05), and significantly promoted the expression of hsa-mi R-218-5 p and hsa-mi R-185-5 p(P<0.05). Conclusion: Heat-clearing and blood-activating components may play a protective role in the brain by promoting the expression of mi RNA/VEGF after acute cerebral infarction and improving the patients’ NIHSS score, namely the symptoms of neurological dysfunction.
