摘  要:目的:探讨经方三物黄芩汤、大黄牡丹汤及薏苡附子败酱散对条件性基因敲除APCMin/+小鼠肠道腺瘤发生和肠道菌群的影响。方法:将30只APCMin/+小鼠随机分为对照组、三物黄芩汤组、大黄牡丹汤组、薏苡附子败酱散组、阿司匹林组,每组6只,对照组给予0.9%氯化钠溶液灌胃,其余4组分别给予对应药物灌胃,于灌胃前、灌胃第12周无菌收集各组小鼠粪便,比较各组小鼠体质量及一般情况,运用16S rRNA高通量测序法检测各组小鼠粪便中微生物群落多样性变化。结果:与对照组比较,各给药组APCMin/+小鼠DAI评分、大便隐血、大便黏度、肠道菌群的OTU数量均有所改善,尤以薏苡附子败酱散组改善效果最为明显;与对照组比较,薏苡附子败酱散组小鼠肠道菌群OUT数量明显增加,其中,乳酸菌属、双歧杆菌属等有益物种呈明显上升趋势,拟杆菌属、norankfErysipelotrichaceae等物种呈下降趋势(P<0.05)。结论:经方薏苡附子败酱散、三物黄芩汤、大黄牡丹汤均能够通过改善肠道菌群结构及多样性,减少APCMin/+小鼠肠道肿瘤数量,改善小鼠的生活质量,其中薏苡附子败酱散效果最好。Objective: To investigate the effects of classical formula on intestinal flora diversity in APCMin/+ mice. Methods: Thirty APCMin/+ mice were randomly divided into 5 groups including control group, Sanwu Huangqin Decoction group(SWHQ), Dahuang Mudan Decoction group(DHMD), Yiyi Fuzi Baijiang Powder group(YYFZBJS) and Aspirin group(Aspirin), with 6 mice in each group. The control group was given normal saline gavage, the other four groups were given the corresponding medicine. The fecal samples of each group were collected before and 12 weeks after gavage. The body weight and general condition of each group were compared. The diversity of microbial community in the feces of each group was detected by 16 S rRNA high-throughput sequencing method. Results: Compared with the control group, the DAI Score, fecal occult blood, fecal viscosity and the number of OTU of intestinal flora of the classical prescriptions were improved, YYFZBJS was the most effective. Compared with the control group, the number of OTU of YYFZBJS was significantly increased, and the beneficial species of lactobacillus and Bifidobacterium of YYFZBJS were significantly increased, bacteroides, norankfErysipelotrichaceae and other species showed a downward trend(P<0.05). Conclusion: YYFZBJS, SWHQ, DHMD can all reduce the number of intestinal tumors in APCMin/+ mice improve the life quality of intestinal adenoma mice by improving the intestinal flora structure and diversity, among these,YYFZBJS is the best.
