摘  要:目的:从周细胞出发,建立周细胞炎症损伤模型,通过研究黄芪甲苷(AS-Ⅳ)在炎性反应中发挥的作用,以期为其应用开发提供科学依据。方法:利用氧化低密度脂蛋白(ox-LDL)体外诱导建立周细胞炎症损伤模型,细胞增殖及毒性检测试剂盒(CCK8)检测不同时间不同浓度的ox-LDL。利用RT-PCR检测c-Jun氨基末端激酶(JNK)、肿瘤坏死因子-α(TNF-α)、核转录因子κB(NF-κB)mRNA表达情况。利用Western blot检测JNK、TNF-α、NF-κB蛋白表达情况。CCK8检测不同浓度的AS-Ⅳ对炎症模型的影响。Transwell法和划线法检测AS-Ⅳ对周细胞趋化功能的影响。流式凋亡细胞仪检测AS-Ⅳ对周细胞凋亡功能的影响。结果:与对照组比较,oxLDL模型组能提高JNK、TNF-α、NF-κB mRNA及蛋白表达水平(P<0.01,P<0.05)。与模型组比较,药物组可以降低JNK、TNF-α、NF-κB mRNA及蛋白表达水平(P<0.05,P<0.01),能提高周细胞的趋化能力(P<0.05),缓解周细胞的凋亡(P<0.05)。结论:AS-Ⅳ具有缓解炎性反应、改善细胞生物学功能的作用。Objective:Starting from pericytes,the inflammation model of pericytes was established,and the role of AstragalosideⅣ(AS-Ⅳ)in inflammation was studied,so as to provide scientific basis for its application and development.Methods:Oxidative low-density lipoprotein(ox-LDL)was used to induce the model of pericytes inflammatory damage model in vitro,and the cell counting kit-8(CCK8)was used to detect ox-LDL at different concentrations and different times.mRNA expression of Jun N-terminal kinase(JNK),tumor necrosis factor-alpha(TNF-α)and nuclear transcription factor kappa B(NF-κB)were detected by RT-PCR.The protein expressions of JNK,TNF-αand NF-κB were detected by Western blot.CCK8 detected the effects of AS-Ⅳat different concentrations on the inflammatory model.Transwell experiment and scratch experiment tested the effect of AS-Ⅳon biological function of pericytes.Flow cytometry was used to detect the effect of AS-Ⅳon apoptosis of pericytes.Results:Compared with the control group,ox-LDL increased the mRNA and protein expression levels of JNK,TNF-αand NF-κB(P<0.01,P<0.05).Compared with the model group,AS-Ⅳcould reduce the mRNA and protein expression levels of JNK,TNF-αand NF-κB(P<0.05,P<0.01),improved the chemotaxis ability of pericytes(P<0.05),and alleviate the apoptosis of pericytes(P<0.05).Conclusion:AS-Ⅳcan alleviate the inflammatory response and improve the biological function of pericytes.
