摘  要:目的:探寻和评估适用于研究糖尿病胃轻瘫(DGP)的动物模型。方法:通过单光子发射计算机断层成像术(SPECT)/计算机断层成像术(CT)测定核素99mTc-DTPA液体胃排空测定法及小肠推进率法对比高脂饲料喂养条件下链脲菌素(STZ)造模SD大鼠(STZ组)及Zucker大鼠(ZDF组)形成DGP模型情况。结果:SPECT/CT扫描结果显示,STZ组和ZDF组造模后均出现稳定的胃排空障碍,ZDF组第46分钟后胃中的残留活性与正常组差异有统计学意义(P<0.05),小肠推进率低提示ZDF组和STZ组的胃排空较慢,这与SPECT/CT扫描的实验结果一致。结论:从胃排空障碍角度来看,ZDF大鼠与人类2型糖尿病患者更为相似,且具有更高的稳定性。ZDF大鼠配合SPECT/CT动态胃排空扫描的研究方法能够保证实验的完整性,可以进一步研究中医疗法治疗DGP的机制。Objective:To explore and assess a suitable animal model for studying acupuncture therapy to treat diabetic gastroparesis(DGP).Methods:A DGP model was developed with streptozotocin(STZ)administration in Sprague-Dawley(SD)rats and Zucker rats fed a high-fat diet(ZDF).Single-photon emission computed tomography(SPECT)/CT scanning with the radionuclide 99mTc-DTPA-based liquid gastric-emptying assay and the small intestinal transit rate were used to assess successful establishment of the DGP model.Results:The SPECT/CT scanning results showed that STZ group and ZDF group had slower gastric movement after modelling.The residual activity in the stomach after the 46th minute in ZDF group was significantly different from that in normal group(P<0.05).The small intestinal transit rate results revealed slower gastric emptying in ZDF group and STZ group,which were consistent with the experimental results of SPECT/CT scanning.Conclusion:ZDF rats more closely resemble patients with type 2 diabetes in terms of gastrointestinal dysfunction during disease development,and ZDF rats have higher stability.ZDF rats combined with dynamic SPECT/CT scanning gastric emptying method can ensure the integrity of the experiment,which can further study the mechanism of traditional Chinese medicine treatment of DGP.
