摘 要:研究杂色云芝粗多糖(YZ0)对对乙酰氨基酚(APAP)诱导的小鼠急性肝损伤的保护作用及其作用机制,结果表明:与模型组相比,YZ0能显著降低小鼠血清ALT、AST、IL-1β、TNF-α、IL-6水平及肝MDA水平,显著增强肝组织中抗氧化酶(SOD、CAT)活性和GSH水平,并显现剂量依赖性。组织病理学观察显示,YZ0对肝损伤有保护作用。此外,YZ0显著抑制了APAP诱导的ERK1/2和JNK的磷酸化,从而减少了促炎细胞因子的分泌和肝细胞凋亡,揭示了YZ0对APAP诱导的小鼠肝毒性具有保护作用,其可能作用机制是减少氧化应激和炎症反应。The present study was designed to investigate the hepatoprotective effect of polysaccharides from Coriolus versicolor(YZ0)and their possible mechanisms in acetaminophen(APAP)-induced acute liver damage in mice.The results indicated that YZ0 significantly decreased serum ALT and AST activities,IL-1β,TNF-αand IL-6 serum levels,hepatic MDA level,and markedly enhanced antioxidant enzyme(SOD and CAT)activity and GSH level in hepatic tissue,in a dose dependent manner,when compared to the APAP group.Histopathological observation revealed the hepatoprotective effect of YZ0 against damage.Furthermore,YZ0 remarkably improved the APAP-induced phosphorylation of ERK1/2 and JNK,therefore to prevent the secretion of pro-inflammatory cytokines and hepatocytes apoptosis.These findings suggested that YZ0 possessed hepatoprotective effect against APAP-induced hepatotoxicity in mice and the action might in part reduce oxidative stress and inflammation.
