摘  要:为验证DNA条形码在水螨类群中的可行性,测定6种水螨9个样本的COⅠ基因序列,并从GenBank中下载我国的4种水螨11条序列,合计10种水螨20条序列.在GenBank和BOLD Systems数据库中对9条序列进行比对和分子鉴定;采用DAMBE软件对20条序列进行碱基饱和度分析;MEGA7.0软件进行碱基序列分析,遗传距离分析以及NJ系统发育树的构建.结果显示:9条序列在GenBank和BOLD中均未能通过分子数据鉴定到具体的种类;20条序列中的A+T含量为66.4%,表现出明显的A-T偏好性,与节肢动物线粒体碱基组成基本相符;碱基替换成线性关系,未达到饱和状态,可用于后续分析;种内最大遗传距离(0.012)小于种间的最小遗传距离(0.158),存在着明显的条形码间隔;在系统发育树中,同一物种的个体均能聚成一支.研究表明基于COⅠ基因的DNA条形码技术能够在种级阶元上对水螨物种进行有效区分.In order to verify the feasibility of DNA barcoding in water mite species,9 sequences(fragments of COⅠgene)of 6 water mite species were sequenced in our study,11 sequences of 4 Chinese species were downloaded from GenBank.The total 20 sequences of 10 water mite species were analyzed in this study.9 sequences were aligned and identified by GenBank and BOLD Systems.The base substitution saturation of 20 sequences were analyzed by DAMBE,the analysis of base composition,the genetic distance and the phylogenetic trees(based on neighbor-joining method)were performed with MEGA7.0.The result showed that none sequneces was identified as species by GenBank and BOLD Systems.The base of A+T content reached up to 66.4%,which consistent with the basic characteristics of mitochondrial gene base composition in arthropod.The base substitution was undersaturated and showed linear relationships.It was proved that the sequences could be used for the subsequent analysis.The barcoding gap was obvious because the minimum inter-specific genetic distance(0.158)was much higher than maximum intra-species genetic distance(0.012).The phylogenetic tree indicated that all the individuals of the same species would cluster in the same clade.Therefore,this study demonstrated that the technology of DNA barcoding based on COⅠis effective for the identification of water mites on species level.
