摘  要:为实现未来中国空间站上微流控芯片中的单线虫自动化培养和生长监测,确定适合线虫生长的液体培养条件和生长过程中的一些关键参数,分析了单线虫控制和长期培养的关键限制因素。通过采取适合的无菌液体培养基进行线虫的驯化,获得了寿命达30天以上的稳定生长线虫,并对线虫幼虫发育至成虫过程中的体长、体宽、产卵指标以及监测时对光照的敏感性进行了分析。获得了适应液体培养基的线虫的体长、体宽发育曲线、产卵期和存活曲线,数据表明:光照对线虫的发育具有抑制作用,且抑制作用因光源而异。研究结果为微流控自动化动物培养和监测的限制条件提供了参数。To realize the automatic cultivation and growth monitoring of single C.elegans in a microfluidic chip onboard the China Space Station in the future,the suitable liquid culture conditions and some key parameters in the growth process of nematodes were determined,and the key limiting factors for the control and long-term cultivation of nematodes were analyzed.By adopting suitable sterile liquid culture medium to domesticate nematodes,stable growth nematodes with a life span of more than 30 days were obtained.The body length and the body width of nematodes during development from larvae to adults,the oviposition index,and the sensitivities to light during monitoring were analyzed.The data curves of the body length,the body width,the oviposition period and the survival rate of nematodes adapted to liquid culture medium were obtained.The results showed that light could inhibit the development of nematodes,and the inhibition was different for different light sources.The results would provide parameters and monitoring limitations for culturing model animal in microfluidic chip automatically.
