摘  要:目的初步探究黄芪多糖(APS)通过自噬途径对链脲佐菌素(STZ)诱导的糖尿病心肌病(DCM)大鼠心肌细胞凋亡的影响。方法将30只SPF级雄性SD大鼠随机分为正常对照组(NC组)、DCM组、APS治疗组(APS组)。DCM组和APS组大鼠一次性腹腔注射10 g/L的STZ 50 mg/kg建立糖尿病模型,NC组大鼠腹腔注射等体积的柠檬酸钠缓冲液;APS组大鼠按1 g·kg^-1·d^-1灌胃APS,其余两组大鼠灌胃等体积生理盐水。干预16周后,采用经胸超声心动图评估大鼠的心功能,苏木精-伊红染色观察心肌组织病理学改变,TUNEL染色检测心肌细胞凋亡情况,Western blot检测心肌组织内自噬相关蛋白p62、LC3的表达水平。结果与NC组相比,DCM组大鼠心功能显著降低(F=5.584~342.701,t=3.125~18.648,P<0.05);心肌组织病理学改变明显,表现为心肌细胞排列紊乱、细胞界限不清、肌纤维断裂等,心肌组织内细胞凋亡增多;LC3蛋白表达显著降低,p62蛋白表达显著增加(F=11.341、22.155,t=4.439、9.409,P<0.05)。APS干预16周后,与DCM组相比,APS组大鼠心功能得到有效恢复(t=2.503~13.416,P<0.05);心肌结构损伤明显减轻,心肌细胞凋亡减少;LC3蛋白表达显著增加,p62蛋白表达显著减少(t=3.714、9.682,P<0.05)。结论APS可能通过上调心肌细胞内自噬水平而抑制STZ诱导的DCM大鼠心肌细胞凋亡,发挥心脏保护作用。Objective To investigate the effect of Astragalus polysaccharides(APS)on cardiomyocyte apoptosis in rats with streptozotocin(STZ)-induced diabetic cardiomyopathy(DCM)via the autophagy pathway.Methods A total of 30 specific pathogen-free male Sprague-Dawley rats were randomly divided into normal control group(NC group),DCM group,and APS treatment group(APS group).The rats in the DCM group and the APS group were given a single intraperitoneal injection of 10 g/L STZ(50 mg/kg)to establish a model of diabetes,and those in the NC group were given intraperitoneal injection of an equal vo-lume of sodium citrate buffer.The rats in the APS group were treated with APS(1 g·kg^-1·d^-1)by gavage,and those in the other two groups were given an equal volume of normal saline by gavage.After 16 weeks of intervention,transthoracic echocardiography was used to evaluate cardiac function,HE staining was used to observe myocardial histopathological changes,TUNEL staining was used to observe cardiomyocyte apoptosis,and Western blot was used to measure the expression of the autophagy-related proteins p62 and LC3 in the myocardium.Results Compared with the NC group,the DCM group had a significant reduction in cardiac function(F=5.584-342.701,t=3.125-18.648,P<0.05)with marked myocardial histopathological changes(disordered arrangement of cardiomyocytes,unclear cell boundaries,and myofibril breakage)and a significant increase in cardiomyocyte apoptosis with a significant reduction in the protein expression of LC3 and a significant increase in the protein expression of p62(F=11.341,22.155;t=4.439,9.409;P<0.05).After 16 weeks of APS intervention,compared with the DCM group,the APS group had effective recovery of cardiac function(t=2.503-13.416,P<0.05),marked alleviation of myocardial structural damage,and a significant reduction in cardiomyocyte apoptosis,as well as a significant increase in the protein expression of LC3 and a significant reduction in the protein expression of p62(t=3.714,9.682;P<0.05).Conclusion APS may inhibit cardiomyoc
