摘 要:目的观察Sprouty 2(Spry 2)和细胞外调节蛋白激酶1/2(ERK1/2)在复合应激勃起功能障碍模型大鼠阴茎组织中的表达及伊木萨克片对二者表达的影响,探讨伊木萨克片的作用机制。方法选用50只正常的雄性Sprag-Dawley大鼠,随机抽取10只作为正常组,余40只大鼠采用环境雌激素样饮食联合冷应激干预条件建立复合应激勃起功能障碍模型。建模24周后,通过阿朴吗啡(APO)勃起实验筛选出勃起功能障碍大鼠20只,然后随机分为模型组和伊木萨克片组各10只。模型组和正常组大鼠给予蒸馏水灌胃,伊木萨克片组大鼠给予伊木萨克片250 mg/kg灌胃,均每天1次,连续2周。干预2周后,免疫组化方法检测3组大鼠阴茎组织中Spry 2和ERK1/2蛋白表达情况。结果模型组大鼠阴茎组织中Spry 2表达水平明显低于正常组(P<0.05),伊木萨克片组大鼠阴茎组织中Spry 2表达水平明显高于模型组(P<0.05);3组大鼠阴茎组织中总ERK1/2(t-ERK1/2)表达水平比较差异无统计学意义(P>0.05);模型组大鼠阴茎组织中磷酸化ERK1/2(p-ERK1/2)表达水平明显高于正常组(P<0.05),伊木萨克片组大鼠阴茎组织中p-ERK1/2表达水平明显低于模型组(P<0.05)。结论环境激素样饮食联合冷应激诱导的Spry 2-ERK1/2通路的活化可促进勃起功能障碍的发生发展,而伊木萨克片可能通过抑制此通路对复合应激勃起功能障碍大鼠模型发挥保护作用。Objective It is to observe the expression of Sprouty 2(Spry 2)and extracellular regulatory protein kinase 1/2(ERK1/2)in the penile tissue of rats with compound stress erectile dysfunction and the effect of Imusak tablets on them,and to explore the action mechanism of this medicine.Methods Fifty normal male Sprag-Dawley rats were selected,in which 10 rats were randomly selected as the normal group.The remaining 40 rats were given environmental estrogen-like diet combined with cold stress intervention to establish the models of compound stress erectile dysfunction.After 24 weeks of modeling,20 rats with erectile dysfunction were screened by the apomorphine(APO)erection test,and then were randomly divided into model group and Imusak tablet group,10 rats in each group.The rats in the model group and the normal group were given distilled water by intragastric administration,and the rats in the Imusak tablet group were given 250 mg/kg Imusac tablet by intragastric administration,all once a day,and continuously treatd for 2 weeks.After 2 weeks of intervention,immunohistochemical methods were used to detect the expression of Spry 2 and ERK1/2 protein in the penile tissues of the three groups.Results The expression level of Spry 2 in the penis tissue of the model group was significantly lower than that of the normal group(P<0.05),and the expression level of Spry 2 in the penis tissue of the Imusak tablet group was significantly higher than that of the model group(P<0.05).There was no significant difference in the expression level of total ERK1/2(t-ERK1/2)in rat penile tissue(P>0.05)among the three groups(P<0.05).The expression level of phosphorylated ERK1/2(p-ERK1/2)in rat penile tissue of model group was significantly higher than that in the normal group(P<0.05),and the expression level of p-ERK1/2 in the penis tissue of the rats in the Imusak tablet group was significantly lower than that in the model group(P<0.05).Conclusion The environmental hormone-like diet combined with cold stress-induced activation of the Spry 2-
