摘 要:目的探讨胃痛消痞方对肝郁脾虚型功能性消化不良(FD)大鼠胃肠动力及5-羟色胺受体(5-HT3aR)表达的影响,探讨其可能的作用机制。方法选用改良郭氏夹尾刺激法联合张氏不规则喂食法复制FD大鼠模型,阳性对照药选用莫沙必利片,中药干预以胃痛消痞方灌胃治疗。实验结束后,观察各组大鼠的一般状态,记录胃内残留率及小肠推进率,WB法观察胃窦及下丘脑组织中5-HT3aR蛋白的变化。结果造模组大鼠一般状态差,胃内残留率增加,小肠推进率降低,胃窦及下丘脑组织中5-HT3aR蛋白表达降低(P<0.05);胃痛消痞方组及西药组大鼠一般状态明显改善,胃内残留率降低,小肠推进率明显升高,胃窦及下丘脑组织中5-HT3aR蛋白表达升高(P<0.05)。结论胃痛消痞方能促进FD大鼠胃肠动力,上调5-HT3aR蛋白表达,可能是其治疗FD的作用机制之一。Objective To observe the effect of Weitongxiaopi prescription on gastrointestinal motility and ex-pression of serotonin receptor(5-HT3aR)in functional dyspepsia(FD)rats with liver depression and spleen deficien-cy,and to explore its possible mechanism.Methods The modified Guo's clip tail stimulation method combined with Zhang's irregular feeding method was used to replicate the FD rat model.The positive control drug was mosapride,and Weitongxiaopi prescription was used for Chinese medicine intervention by gavage.After the experiment,the general state of rats in each group was observed,the intragastric residual rate and intestinal propulsive rate were recorded,and the changes of 5-HT3aR protein in the gastric antrum and hypothalamus tissue was observed by WB method.Results The general state of rats in model group was poor,the intragastric residual rate was increased,while the intestinal pro-pulsive rate was decreased,and the 5-HT3aR protein expression in the gastric antrum and hypothalamus tissue was de-creased(P<0.05);the general state of rats in Weitongxiaopi group and western medicine group was significantly im-proved,the intragastric residual rate was decreased,while the intestinal propulsive rate was significantly increased,and the 5-HT3aR protein expression in the gastric antrum and hypothalamus tissue was increased(P<0.05).Conclusion Weitongxiaopi prescription can promote gastrointestinal motility of FD rats and increase the expression of 5-HT3aR pro-tein,which may be one of the mechanisms of its treatment for FD.
