摘 要:目的探究Nesfatin-1对高糖高脂诱导的睾丸间质细胞(TM3细胞)氧化应激的干预作用。方法培养体外小鼠睾丸间质细胞(TM3细胞),采用25 Mm的葡萄糖(Glucose)与62.5 Um的油酸(OA)诱导6 h、12 h、24 h建立细胞模型,分为5组:对照组(Control组,C组)、高糖模型组(G组)、高脂模型组(O组)、高糖高脂模型组(GO组)、高糖高脂+Nesfatin-1(10-8M)干预组(GO+N干预组),通过CCK-8检测细胞存活率,并测定细胞内超氧化物歧化酶(SOD)的含量和丙二醛(MDA)的含量。结果与对照组比较,高糖组、高脂组和高糖高脂组TM3细胞在12~24 h中,细胞抑制率逐渐升高,存活率降低(P均<0.05),高糖组、高脂组和高糖高脂组,TM3细胞内SOD活力降低,MDA水平升高(P均<0.05);与高糖高脂组相比较,高糖高脂组+Nesfatin-1干预组可提高高糖高脂状态下细胞的存活率,逆转存活率的降低,并可提升细胞内SOD活性、降低TM3细胞内的MDA水平。结论 10-8M Nesfatin-1可逆转因高糖高脂所诱导的TM3存活率的降低,发挥明显的抗氧化应激作用。Objective To investigate the protective role of Nesfatin-1 on oxidative stress in Leydig cells(TM3)induced by high glucose and high fat.Methods Mouse TM3 cells were cultured in vitro.TM3 cells were induced by 25 Mm glucose and 62.5 Um oleic acid(OA)for 6 h,12 h,and 24 h respectively.They were randomly divided into seven groups:control group(C group),glucose group(G group),oleic acid group(O group),glucose and oleic acid group(GO group),glucose Nesfatin-1 group(10-8M)(G+N group),oleic acid Nesfatin-1 group(10-8 M)(O+N group),glucose and oleic acid Nesfatin-1 group(10-8M)(GO+N group).After treatment by 10-8M Nesfatin-1 for 1 h,the effect of Nesfatin-1 on oxidative stress of TM3 cells,induced by high glucose and high fat was detected by CCK-8.The contents of SOD and MDA in cells were measured by colorimetric method.Results Compared with the control group,the cell suppression rate of TM3 cells in the G group,O group,GO group increased gradually and the survival rate decreased significantly(P all<0.05),the SOD activity in TM3 cells decreased and the MDA level increased in the G group,O group,GO group,compared with the GO group(P all<0.05),GO+N group can improve the survival rate of cells under high glucose and high fat state,reverse the decrease of survival rate,and can increase the intracellular SOD activity and reduce the MDA level in TM3 cells.Conclusion 10-8M Nesfatin-1 can reverse the decrease of the survival rate of the testicular interstitial cells(TM3)induced by high glucose and high fat,and play an significant anti-oxidative stress role.
