摘  要:以小鼠作为模型动物,观察和验证豆粕当中的抗营养因子α-伴大豆球蛋白刺激小鼠中性粒细胞(PMNs)之后,是否能诱导其释放胞外诱捕网(NETs),并初步揭示其作用机制,从而为深入研究豆粕当中抗营养因子的作用机制提供参考。通过分离小鼠外周血PMNs,培养液中加入α-伴大豆球蛋白孵育后,激光共聚焦进行形态观察,Western blot检测细胞MAPK信号转导通路下游ERK和p38蛋白表达的影响,同时检测培养液中的乳酸脱氢酶(LDH)。结果表明,α-伴大豆球蛋白能够刺激小鼠PMNs释放网状NETs结构,其上装饰有精氨酸、髓过氧化物酶等活性成分,NETs释放水平和加入的α-伴大豆球蛋白剂量呈正相关,且ERK和p38通路可能在α-伴大豆球蛋白诱导NETs释放过程中发挥作用,而LDH水平未见升高。以上结果证实豆粕当中的抗营养因子α-伴大豆球蛋白,刺激小鼠PMNs之后可诱导其释放NETs。α-conglycinin is one of the components of glycinin,also acts as an antigenic protein,which causes an immune response when it enters the body.As the first line of defense of the body,neutrophils play an important role in the early stages of pathogen invasion.Neutrophils play an anti-infective role through phagocytosis and degranulation.In recent years,the neutrophil extracellular trapping network has also played a defense role in microbial invasion.At present,the mechanism of α-conglycinin-induced extracellular trapping of neutrophils has not been reported.In this study,laser confocal microscopy and western blotting were used to investigate the effect of α-conglycinin on the formation of extracellular trapping network of neutrophils and its potential mechanism.The results showed that α-conglycinin could induce the formation of extracellular trapping network of neutrophils,increase the production of reactive oxygen species,and promote the expression of ERK and p38 in MAPK pathway.The above results show that α-conglycinin can induce the extracellular trapping network of neutrophils,which lays a good foundation for further study on the influence and mechanism of α-conglycinin on the immune system.
