健脾化痰方对高脂血症脾虚痰浊证模型小型猪血管内皮功能的影响

摘　　要：目的探讨健脾化痰方治疗高脂血症脾虚痰浊证的可能作用机制。方法普通级广西巴马小型猪15只随机分为正常组、模型组和给药组,每组5只。正常组予基础饲料喂饲,模型组和给药组均予高脂饲料喂饲24周建立高脂血症脾虚痰浊证小型猪模型。给药组在高脂饲料喂饲同时给予健脾化痰方颗粒剂,将颗粒剂溶于100ml生理盐水中,均匀掺入饲料内,每日1剂,喂饲24周。在0周和24周时采集小型猪空腹耳静脉血检测血清内皮功能指标包括一氧化氮(NO)、内皮型一氧化氮合酶(eNOS)、诱导型一氧化氮合酶(iNOS)、总一氧化氮合酶(tNOS),及血清内皮素1(ET-1)、细胞间黏附分子1(ICAM-1)水平;24周时检测冠状动脉内皮组织匀浆中内皮功能指标及ET-1、ICAM-1水平。结果24周时,与对照组比较,模型组小型猪血清和冠状动脉内皮组织匀浆中tNOS、iNOS、ET-1、ICAM-1水平均升高,eNOS、NO水平降低(P<0.05);与模型组比较,给药组小型猪血清和冠状动脉内皮组织匀浆中tNOS、iNOS、ET-1、ICAM-1水平均降低,eNOS、NO水平升高(P<0.05)。结论健脾化痰方能有效改善血管内皮功能,可能是其治疗高脂血症脾虚痰浊证的作用机制之一。Objective To explore the possible mechanism of Jianpi Huatan Formula(健脾化痰方)in the treatment of hyperlipidemia with spleen deficiency and phlegm turbid syndrome.Methods A total of 15 ordinary Guangxi Bama minipigs were randomly divided into a normal group,a model group and a treatment group,with 5 in each group.The normal group was fed with basic feed,the model group and the treatment group were fed with high-fat feed for 24 weeks to establish a small pig model of hyperlipidemia with spleen deficiency and phlegm turbidity syndrome.The treatment group was given Jianpi Huatan Formula Granules(健脾化痰方颗粒剂)at the same time given high-fat feed.The granules were dissolved in 100 ml of normal saline and evenly mixed into the feed.One dose per day for 24 weeks.At 0 week and 24 weeks,fasting auricular venous blood of the minipigs was collected to detect the levels of nitric oxide(NO),endothelial nitric oxide synthase(eNOS),inducible nitric oxide synthase(iNOS),total nitric oxide synthase(TNOs),serum endothelin 1(ET-1)and intercellular adhesion molecule 1(ICAM-1).After 24 weeks,the endothelial function index and ET-1,ICAM-1 level in endothelium homogenate of coronary artery was measured.Results At 24 weeks,compared with the control group,the levels of tNOS,iNOS,ET-1,ICAM-1 were increased,and the levels of eNOS,NO were decreased in the model group(P<0.05).Compared with the model group,the levels of tNOS,iNOS,ET-1,ICAM-1 in minipig serum and coronary artery endothelial tissue homogenate were decreased,and the levels of eNOS and NO were increased(P<0.05).Conclusion Jianpi Huatan Formula(健脾化痰方)can effectively improve the function of endothelium,which may be one of the mechanisms of its treatment of hyperlipidemia with spleen deficiency and phlegm turbid syndrome.
