伊犁马PITX2基因多态性及其与产奶性状的关联分析

摘　　要：为了研究伊犁马垂体同型框转录因子2(paired-liked homeodomain transcription factor 2,PITX2)基因的多态性及其与产奶性状间的关系,试验采用PCR-SSCP和DNA测序技术,对80匹伊犁马PITX2基因的多态性进行研究,并利用SPSS 18.0软件中的General Linear Model(GLM)模型分析了PITX2基因与产奶性状的关系。结果表明:PITX2基因在乳用型伊犁马中存在多态性,在内含子区域g.115289715处缺失碱基A,产生三种基因型,即AA、AB和BB,经关联分析发现这三种基因型个体的产奶性状间存在显著关联。说明PITX2基因的多态位点有可能成为伊犁马产奶性状选育的分子标记。In order to study the polymorphism of the pituitary isoform transcription factor 2(PITX2)gene in Yili horse and its relationship with milk production traits,the polymorphism of PITX2 gene in 80 Yili horses was studied by using PCR-SSCP and DNA sequencing,The relationship between the PITX2 gene and milk production traits was analyzed using the General Linear Model(GLM)in the software SPSS 18.0.The results showed that PITX2 gene was polymorphic in dairy Yili horses,and the deletion of base A in intron region g.115289715 resulted in 3 genotypes,namely AA,AB and BB.The correlation analysis revealed that there was significant correlation between milk traits of three different genotypes.The results indicated that the polymorphic locus of PITX2 gene might be used as a molecular marker for breeding traits in Yili horses.
