摘 要:[目的]比较免疫抑制模型大鼠脑脊液中两种新型隐球菌感染的检测方法的检出率.[方法]将60只雄性大鼠,随机分为A、B、C、D组,A组和C组大鼠实验d1一次性腹腔注射环磷酰胺(150 mg/kg),B组和D组腹腔注射等量1 m L生理盐水;实验d4 A组和B组大鼠尾静脉注射1 m L浓度为1×106新型隐球菌悬液,建立大鼠感染模型,C组和D组大鼠注射1 m L生理盐水作为阴性对照.实验d6收集四组大鼠的脑脊液,同时采用荧光定量聚合酶链式反应法(FQ-PCR)和墨汁染色方法检测四组大鼠脑脊液,比较两种方法的新型隐球菌感染阳性检出率.[结果]新型隐球菌FQ-PCR方法检测新型隐球菌感染阳性检出率高于墨汁染色方法,其差异有统计学意义(P<0.05).[结论]FQ-PCR检测大鼠脑脊液中新型隐球菌感染阳性率优于墨汁染色,可用于临床检测脑脊液中是否存在新型隐球菌感染.[Objective]Real‐time fluorescent quantitative PCR(FQ‐PCR)and ink staining were used to de‐tect rat cerebrospinal fluid(CSF)in a rat model of Cryptococcus neoformans infection.The sensitivity and spe‐cificity of the two methods were compared to provide laboratory basis for clinical application.[Methods]Sixty male rats were randomly divided into four groups:group A and group C were injected with cyclophosphamide(150 mg/kg)once on the first day of the experiment,while group B and group D were injected with the same amount of 1 mL saline intraperitoneally.To establish infection model,rats in group A and group B were injec‐ted with 1 mL(containing 1×106)of Cryptococcus neoformans suspension in the tail vein on the fourth day of the experiment,while rats in group C and group D received 1 ml saline as negative control.On the 6th day of the experiment,the cerebrospinal fluid of all rats was collected.Then Cryptococcus in cerebrospinal fluid of the four groups was detected by FQ‐PCR and stained with ink at the same time.The results from the two methods were compared.[Results]The positive detection rate of Cryptococcus neoformans by FQ‐PCR was higher than that of ink staining,and the difference was statistically significant(P<00.5).[Conclusion]FQ‐PCR system is superior to ink staining in detecting Cryptococcus neoformans in cerebrospinal fluid of rats.It can be used for clinical detection of the presence of Cryptococcus neoformans infection in cerebrospinal fluid.
