EXAKT硬组织切磨技术在含有植入物组织切片中的应用

摘　　要：目的用EXAKT硬组织切磨技术制作不同类型的软硬组织切片,探索该技术在不同类型组织中的应用及其效果。方法取含有钛合金螺纹种植钉的比格犬下颌骨组织(A组织)、含有钛合金直钉和螺纹种植钉的小鼠股骨组织(B组织和C组织)、含有多孔钛材料的兔股骨踝骨组织(D组织)、含有钴铬合金支架的兔心血管组织(E组织)、含有钙黄绿素和茜素络合复合物标记的大鼠上腭组织(F组织),均经多聚甲醛固定、乙醇上行脱水、光固化树脂浸润包埋后,用EXAKT硬组织切片系统粗切为厚片,再经磨片系统打磨至50μm左右的薄片,染色封片后在光学显微镜下进行观察。结果Ladewig染色后镜下观察显示,A和B组织中骨组织与种植钉结合紧密,种植钉无脱落,新生骨明显;苏木精-伊红染色后镜下观察显示,C和D组织中新生骨明显,与种植体结合紧密,结缔软组织保留完整,原旧骨无裂痕;E组织血管壁清晰可见,分层明显,支架无脱落;荧光显微镜下观察,F组织红绿荧光明显可见,叠加图可清晰看到荧光走向。结论EXAKT硬组织切磨系统可将不同类型的软硬组织制作成切片,且植入物无脱落现象,骨组织无裂痕,各种软硬组织及植入物清晰可见。Objective To prepare different types of soft and hard tissue sections using EXAKT cutting and grinding system,and to investigate the application and effect of this technique in different types of tissue.Methods The samples of Beagle canine mandible with titanium alloy screw implant(tissue A),mouse femur with titanium alloy straight implant and screw implant(tissue B and tissue C),rabbit femur and anklebone with multihole titanium alloy implant(tissue D),rabbit cardiovascular tissue with Co-Cr stent implant(tissue E),and rat palate labeled by fluorexon-alizarin complex(tissue F)were collected.These samples were fixed with polyformaldehyde,dehydrated with ethanol,and then embedded with UV-curable resin.The EXAKT cutting and grinding system was used to cut the embedded samples into thick sections and grind them down to thin slices with a thickness of about 50μm.The thin sections were stained,sealed,and then observed under a light microscope.Results Ladewig staining showed that in tissue A and B,bone tissue was tightly bound to the screw implant,the screw did not fall off,and new bones were observed.Hematoxylin and eosin staining showed that in tissue C and D,new bones were observed and were tightly bound to the implant,the connective tissue remained intact,and no cracks were observed in the old bone.Tissue E had clear vascular walls with obvious stratification and the stent did not fall off.The fluorescence microscope showed that tissue F had clearly visible red and green fluorescence,and fluorescence trend was observed through multiple images.Conclusion The EXAKT cutting and grin-ding system can be used to prepare the sections of different types of soft and hard tissue,with no removal of implant or cracks on bone tissue,and the hard and soft tissue and implant are clearly visible.
