摘  要:为研究转录因子LFY2的表达和功能,本文以拟南芥为材料研究了LFY2定位和表达特性。使用GFP融合蛋白技术分析LFY2的亚细胞定位,结果显示LFY2定位在细胞核中;定量PCR检测拟南芥不同组织及不同发育时期花中LFY2的相对表达量,结果显示LFY2在花苞中表达量最高;原位杂交检测显示LFY2在拟南芥花药发育的第7时期在绒毡层中开始表达,并在7~12时期持续表达,说明LFY2在花药绒毡层的发育过程中起一定作用。该研究为进一步探究LFY2在花药发育中的功能提供了一定的实验数据。For investigating the expression and function of transcription factor LFY2,this research showed the localization and expression characteristics of LFY2 in Arabidopsis thaliana.The subcellular localization of LFY2 in Arabidopsis was determined by fusing their full-length coding sequences upstream of GFP under the control of the 35S promoter.The GFP signal of LFY2-GFP was detected in the nucleus.The expression of LFY2 in different tissues and flowers of different developmental stages was analyzed by qRT-PCR.The results showed that LFY2 was the highest expression in the flower buds.The expression of LFY2 in the anther was also detected using in situ hybridization.The beginning expression of LFY2 in the Arabidopsis tapetum was at the 7th stage,and rapid decreased at 12th stage.These results indicated that LFY2 might have a role in the tapetum development.Our findings provide valuable insights into the mechanism for the function of LFY2 in anther development.
